Hsd:Athymic Nude-Foxn1nu female mice (6 weeks of age) for xenograft experiments were purchased from Envigo. Mice were housed in a controlled environment (12-h light/12-h dark cycle, light intensity ~350 lux) with ad libitum access to water and standard food (13.5% kcal from fat, LabDiet 5L0D). All animal experiments were performed in compliance with the NIH Guide for the Care and Use of Laboratory Animals and Animal Welfare Act. All animal studies were approved and performed in accordance with the Vanderbilt Institutional Animal Care and Use Committee.

Nude Mice Xenograft Tumor Model

All animal experiments were conducted under a UT-Southwestern IACUC approved protocol. Hsd:Athymic Nude-Foxn1nu female mice were purchased from Envigo. At six to eight-weeks of age (23–25 grams) mice were randomly assigned to the indicated groups. Sample size was determined by the formula: n = 1+2C(s/d)2, in which C = 10.51, based on a power of 90% and a confidence interval of 95%, S = standard deviation of tumor volume (20%), D = tumor burden difference to be considered significant (50%). Therefore, five mice per condition were tested. 2 × 106 cells in 100 µl PBS were subcutaneously injected in the flank. Once tumors were visible, tumor volume was measured by caliper in two dimensions, and volumes were estimated using the equation V = length x width2/2. Caliper measurements were made twice a week.

Xenograft Tumor Induction in Athymic Mice
