Annexin 5 fitc pi apoptosis assay kit

Manufactured by Sungene Biotech
Sourced in China

The Annexin V-FITC/PI apoptosis assay kit is a laboratory tool used to detect and quantify apoptosis, a process of programmed cell death. It utilizes fluorescent markers, Annexin V-FITC and propidium iodide (PI), to identify cells undergoing early and late stages of apoptosis.

Annexin V-FITC/PI Apoptosis Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
The Annexin V-FITC/PI apoptosis assay kit (Sungene Biotech, China) and flow cytometry were employed to analyze apoptosis. Annexin V has a high affinity for phosphatidylserine (PS), which is located in the cell membrane and becomes exposed in the early stage of apoptosis. Flow cytometry was performed with NovoExpress (ACEA Biosciences, USA) according to the ACEA NovoExpress Software Guide. Annexin V+PI– cells were those in the early stage of apoptosis, and Annexin V+PI+ cells were those in the late stage of apoptosis. In our study, the rate of cell apoptosis was represented as the percentage of early apoptotic + late apoptotic cells.

Apoptosis Detection in Kidney Tissue

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cell apoptosis was detected using an Annexin V-FITC/PI apoptosis assay kit (Sungene Biotech, China). In brief, after double-staining with annexin V and propidine iodide (PI), the number of apoptotic cells was measured by flow cytometry analysis. To assess the extent of renal apoptosis, terminal deoxynucleotidyl transferase dUTP nick end-labeling (TUNEL) staining (In Situ Cell Death Detection Kit, POD, Roche) was performed on frozen kidney sections, and the number of TUNEL-positive cells per 0.25 mm2 was calculated.

Detecting Podocyte Apoptosis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Apoptosis was determined using the Annexin V-FITC/PI apoptosis assay kit (Sungene Biotech, China). Flow cytometry with a BD FACSVantage SE cytometer was used to detect the apoptosis rate of cells. Annexin V-positive/PI-negative podocytes were considered to indicate the early stages of apoptosis, whereas annexin V-positive/PI-positive podocytes were considered to be late apoptotic or necrotic cells.

Cytotoxicity and Apoptosis Assay of BSA and PA

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cytotoxicity was estimated by a Cell Counting Kit-8 (CCK-8) colorimetric assay (Sigma-Aldrich, St. Louis, MO, United States). HK-2 cells were seeded onto a 96-well plate and treated with various concentrations of BSA or PA (Sigma, United States, P9767) (0, 100, 150, 200, 250, 300, 350, 400, 450 μM) for 24 h. Then, the treated cells were incubated with a CCK-8 kit (100 μL/well) for 2 h at 37°C in a 5% CO2 incubator. After the plate was agitated, the absorbances of each well at 450 nm were measured using a multiwell fluorescence plate reader (Thermo Scientific Varioskan Flash).
To evaluate the extent of cell apoptosis, an Annexin V-FITC/PI apoptosis assay kit (Sungene Biotech, Tianjin, China) was used to assess cell apoptosis. Briefly, after the indicated treatments, the cells were stained with 10 μg/ml Annexin V/FITC for 30 min at 37°C and with 5 μg/ml propidium iodide (PI) for 5 min at room temperature, followed by flow cytometry analysis using a BD FACSVantage SE cytometer.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
