Metronidazole (MTZ, Cat #M1547, Sigma-Aldrich, St. Louis, MO, USA) was dissolved in embryo medium containing 0.2% DMSO to yield a final concentration of 10 mM as described previously11 (link). Tacrine hydrochloride (Cat #1648–40-8, Tocris, Illkirch, France) and (S)-duloxetine hydrochloride (Cat #136,434–34-9, Tocris) were dissolved in distilled water to yield a 10-mM stock solution. For cell ablation, NTR-transgenic larvae were exposed to 10 mM MTZ for 2 days from 5 to 7 dpf. To test remyelination, MTZ was subsequently washed out using embryo medium, and larvae were incubated in embryo medium containing tacrine, duloxetine, or vehicle (recovery group) for 1 day (Supplementary Fig. 1) prior to analyses.

Fluoxetine, citalopram, prazosin, and reboxetine were dissolved in 1% DMSO. DSP-4, phenylephrine, atomoxetine, propranolol, and duloxetine were dissolved in saline (0.9% NaCl).
