Blood samples were collected in Greiner Bio-One Hematology K3EDTA Evacuated Tubes (Thermo Fisher Scientific). Complete blood counts, including leukocyte differentials and reticulocyte counts (CBC/Diff/Retic), were determined using the Procyte DX (IDEXX Laboratories, Westbrook, ME, USA). The Catalyst One analyzer (IDEXX Laboratories) was used for biochemical analyses of serum samples, which were collected in Greiner Bio-One VACUETTE Z Serum Sep Clot Activator Tubes (Thermo Fisher Scientific). Samples were run on both machines the day of collection shortly following collection such that they were not stored prior to analysis. On D30, an equipment issue with the Procyte DX required that samples from the virus-exposed group be stored at 4°C for about 30 h prior to analysis. Blood Sample Collection and Analysis

At the end of the study, blood samples were collected from the two groups between 19:00 and 19:30. The blood samples were collected in a sterilized tube after cardiac puncture (Kube et al., 2021 (link)). All the samples were placed on ice for 60 min for blood clotting and then centrifuged at 3,000 × g for 10 min, 4°C. The serum samples were then transferred into sterilized tubes for the biochemistry assay. Serum biochemistry profiles, including electrolytes, glucose, amylase, and lipase, were measured using a preloaded Chem 17 CLIP using a Catalyst One analyzer (IDEXX Laboratories, Inc., ME, United States) following the manufacturer’s instructions.

Multiparametric Blood Biomarker Analysis

Blood plasma collected in lithium heparin tubes was used for blood chemistries. The IDEXX Catalyst One Analyzer (IDEXX, Westbrook, Maine, USA) clips included creatinine (CREA), creatine kinase (CK), alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate (LAC), and lactate dehydrogenase (LDH). The custom kit included interleukin (IL)-1β, IL-6, inflammatory protein-10 (IP-10), and leptin.

Serum Biomarkers in Liver Disease

Broiler Chicken Blood Analysis

Red blood cells, white blood cells, lymphocytes, heterophils, monocytes, eosinophils, basophils, platelets, haemoglobin, and other haematological parameters were analysed using an automated IDEXX LaserCyte Hematology Analyzer (IDEXX Laboratories, Westbrook, USA) whilst sero-biochemical parameters (glucose, symmetric dimethylarginine (SDMA), phosphate, calcium, total protein, albumin, globulin, albumin/globulin, alanine transaminase, alkaline phosphatase, total bilirubin, cholesterol, amylase, and lipase) were analysed using an automated IDEXX Catalyst One Analyzer (IDEXX Laboratories, Westbrook, USA).
