Characterization of Shrimp Spoilage Bacteria

The shrimp (L. vannamei) was bought from Qidong Rd. Market, Qingdao, with a weight of 45 ± 5 g. The shrimp was transported to the laboratory on ice within 30 min.
The wild strain of L. plantarum SS-128 and the luxS-mutant strain of L. plantarum SS-128 (ΔluxS) were constructed and preserved by the Laboratory of Aquatic Products Higher Application Technology, Ocean University of China. Shewanella baltica is a laboratory-preserved strain previously isolated from spoiled L. vannamei (Zhu et al., 2015 (link)). DPD was obtained from Professor Ming Li’s lab in the College of Pharmacy, Ocean University of China. The De Man, Rogosa and Sharpe (MRS) broth, Lactobacillus medium, and agar were bought from Qingdao Haibo Biotechnology Co., Ltd., China. Na2HPO4⋅12H2O, NaH2POx⋅H2O, NaCl, 40% Acr-Bis, Tris–HCl, N,N,N′,N′-tetramethylethylenediamine, Tris, dithiothreitol, and other analytical pure chemical reagents were purchased from Sinopharm Group Chemical Reagent Co., Ltd.

Enzymatic Assay for Peroxidase Activity

Expression of Pectate Lyase PEL3 in E. coli

Escherichia coli Rosetta (DE3)/pET28a-pel3 for the expression of pectate lyase PEL3 was constructed in our previous work (Wang et al., 2018 (link)). Glycine with purity 98.5% and polygalacturonic acid (PGA) with purity 90% were purchased from Sigma-Aldrich (St. Louis, MO, United States). CuCl2, ZnSO4, MnSO4, CoCl2, CaCl2, KCl, NaCl, Na2HPO4, KH2PO4, Tris–HCl, and NaOH were purchased from Sinopharm Chemical Reagent Co., Ltd. All above chemicals were analytical grade. PBS buffer (10 mM) was prepared as follow: 4.00 g of NaCl, 0.10 g of KCl, 0.72 g of Na2HPO4, and 0.12 g of KH2PO4 were dissolved in 500 mL deionized water.

(Shanghai, China). All the ODNs were prepared with Tris-HCl buffer (10 mM Tris, 100 mM NaCl, pH 7.4). All reagents were of analytical grade and used as received. Ultrapure water was obtained from a Millipore water purification system (≥18 MΩ, Milli-Q, Millipore).

Degradation Evaluation of MCS-15 in Tris-HCl Buffer

The degradation property of MCS-15 was evaluated at 37°C in 0.05 M Tris(hydroxymethyl) aminomethane-HCl buffer solution (pH 7.4) (Tris-HCl; Sinopharm Chemical Reagent Co., Ltd.,). The MCS-15 was immersed in the buffer solution in the vials. The vials were then loaded onto an orbital shaker with the speed set at 120 rpm. At the scheduled time, the MCS-15 was separated from the solution by centrifugation, washed, and dried at 120°C to a constant weight. The relative weight loss percentage of the samples was calculated by:
