Characterization of Murine Pancreatic Tumor Clones

The clonal cell lines described (E, H, V, K, M, N and T) were isolated from pancreatic tumors from KPC mice and have been previously described30 . Clonal lines 6419c5, 6694c2, 2838c3 and 6499c4 were derived from KPC mice and have also been previously described24 (link). KPC7940B cells were a gift from G. Beatty (University of Pennsylvania). KPC-MT3 cells were a gift from D. Tuveson (Cold Spring Harbor Laboratory). PATC53 cells were obtained from ATCC. Cells were maintained in high-glucose DMEM (Gibco) supplemented with 10% fetal bovine serum (Corning) and routinely tested for mycoplasma contamination using MycoAlert Plus (Lonza). DMSO, oligomycin, phenformin and aminooxyacetic acid were obtained from Sigma, trans-ISRIB was obtained from Cayman Chemical and GCN2iB was obtained from MedChemExpress. IACS-10759 was obtained from Selleck Chem. KPC clonal lines were genotyped for KrasG12D recombination using primer sets from Jackson Laboratories.

Cellular stress response regulation assay

Oxidative Stress and GCN2 Pathway in Diabetic Mice

Compound Library Screening for Bioactive Inhibitors

An 1,833-member bioactive compound library, and an independent 86-member PI3K signaling inhibitor library comprising mTOR, PI3K and AKT pathway inhibitors, were obtained from Selleck Chemicals and stored at −80°C. Erastin2, annotated as compound 35MEW28 (Ref.51 (link)) and ML162 were synthesized by Acme Bioscience. Erastin was the kind gift of B. Stockwell (Columbia). Chemical used (supplier) were: DMSO, ferrostatin-1, thapsigargin, tunicamycin, cycloheximide, L-arginine, D-arginine and L-citrulline (Sigma-Aldrich Corporation); bortezomib, rapamycin, etoposide and buthionine sulfoximine (Thermo-Fisher Scientific); INK 128, AZD8055, vinblastine, camptothecin, sorafenib, bazedoxifene, raloxifene and JTC-801 (Selleck Chemicals); and GCN2iB (MedChemExpress). buthionine sulfoximine was dissolved directly into cell media. All other drugs were prepared as stock solutions in DMSO. Stock solutions were stored at −20˚C.

Evaluation of Oxidative Stress Markers

Modulating Embryonic Protein Translation

Dechorionated embryos were treated with 1 μM SU5416 (Merck KGaA; 676487), 20 μM GSK2656157 (MedChemExpress; HY-13820), 10 μM GCN2-IN-1 (MedChemExpress; HY-100877), 10 μM GCN2iB (MedChemExpress; HY-112654), or 50 μM 4-Phenylbutyric acid (4-PBA) (Sigma Aldrich; P21005) from 24 to 54 hpf. To inhibit global protein translation, the embryos were treated with 360 μM CHX (Sigma-Aldrich; C7698). While 4-PBA was dissolved in water, the other chemicals were dissolved in Dimethyl sulfoxide (DMSO) (Sigma-Aldrich; D8418). The solvents, water or DMSO, were used as controls, respectively, and the final concentration of DMSO is 0.1%.
