Characterization of Shrimp Spoilage Bacteria

The shrimp (L. vannamei) was bought from Qidong Rd. Market, Qingdao, with a weight of 45 ± 5 g. The shrimp was transported to the laboratory on ice within 30 min.
The wild strain of L. plantarum SS-128 and the luxS-mutant strain of L. plantarum SS-128 (ΔluxS) were constructed and preserved by the Laboratory of Aquatic Products Higher Application Technology, Ocean University of China. Shewanella baltica is a laboratory-preserved strain previously isolated from spoiled L. vannamei (Zhu et al., 2015 (link)). DPD was obtained from Professor Ming Li’s lab in the College of Pharmacy, Ocean University of China. The De Man, Rogosa and Sharpe (MRS) broth, Lactobacillus medium, and agar were bought from Qingdao Haibo Biotechnology Co., Ltd., China. Na2HPO4⋅12H2O, NaH2POx⋅H2O, NaCl, 40% Acr-Bis, Tris–HCl, N,N,N′,N′-tetramethylethylenediamine, Tris, dithiothreitol, and other analytical pure chemical reagents were purchased from Sinopharm Group Chemical Reagent Co., Ltd.

Fabrication of Porous Hydrogel Scaffolds

Materials: m-acrylamido phenylboronic acid, and the structure was as follow:

N-[Tris(hydroxymethyl)methyl]acrylamide was purchased from Alfa Aesar, and the structure as follow:

Acrylamide (AAm) was purchased from J&K. Ammonium persulfate and N,N,N′,N′-tetramethylethylenediamine were purchased from Sinopharm. Diphenyliodonium nitrate was purchased from Aladdin.

Preparation: 446 mg acrylamide, 60 mg m-acrylamido phenylboronic acid and 55 mg N-[Tris(hydroxymethyl)methyl]acrylamide were dissolved in the 5 ml deionized water. After fully dissolved, 100 μL Ammonium persulfate and 10 μL N,N,N′,N′-tetramethylethylenediamine were added into the solution. The solution was transferred to a sealed glass vessel quickly. 24 hours later, the resulting gel was immersed in a solution which contains the diphenyliodonium nitrate (control the pH=10) for 24 hours. The gel was frozen by a cooling stage. Under the freezing condition, the gel was exposed by UV irradiation integrally for 5 min. After thawing, the gel swelled in the water to obtain porous gels. The laser scanning confocal microscope was applied to characterize the pore structure of the porous gel.

Shrimp Probiotic and Quorum Sensing Modulation

Shrimp (Litopenaeus vannamei) weighing 45 ± 5 g were purchased from the local fishery market at Qidong Rd., Qingdao, and were transported to the laboratory on ice within half an hour.
Lactobacillus plantarum SS-128 (L. plantarum SS-128) was extracted from the intestine of blackhead fish and preserved by the Laboratory of Aquatic Products Higher Application Technology, Ocean University of China. The luxS-mutant strain of Lactobacillus plantarum SS-128 (L. plantarum ΔluxS) was constructed previously in the lab. Shewanella baltica A02 (S. baltica A02) was isolated from spoiled shrimp. Pro donated the 4,5-dihydroxy-2,3-pentanedione (DPD),Ming Li’s lab in the College of Pharmacy, Ocean University of China. The MRS broth, agar, and Lactobacillus medium were purchased from Qingdao Haibo Biotechnology Co., Ltd., China. The NaCl, N,N,N’,N’-tetramethylethylenediamine, Na2HPO4·12H2O, and other analytical pure chemical reagents were purchased from Sinopharm Group Chemical Reagent Co., Ltd., Shanghai, China.

Synthesis and Characterization of Hydrogel Composites

CMC (CP, viscosity: 300-800 mpa·s) and AM (CP) were purchased from Shanpu Chemical Factory (Shanghai, China) and used without further treatment. Ammonium persulfate (APS, AR), N,N,N'N'-tetramethyl ethylenediamine (TMEDA, AR), Methyl violet (MV), and Methylene blue (MB) were provided by Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China). ATP was obtained from Huangnishan Mine in Xuyi county, Jiangsu province, China, and then ground and passed through a 200-mesh sieve. N,N'-methylenebisacrylamide (MBA, CP) was received from Yuanfan Vegetable Additives (Shanghai, China). T-20 (AR) was received from BASF Corporation. 3A molecular sieve 103 was obtained from Molsion Molecular Sieve Co., Ltd. (Shanghai, China). Linseed oil was purchased from Yongfan Trading Co. (Lanzhou, China). Colza oil and soybean oil were purchased from Yihaijiali Arawana Edible Oil Co., China. Peanut oil, sunflower oil, olive oil, and sesame oil were obtained from Shandong Luhua Group Co., Ltd. China. Corn oil was purchased from Cofco Food Marketing Co., Ltd. China. Other reagents were all of analytical grade, and all solutions were prepared with deionized water.
