Pharmacological Modulation of Vascular Function

hydrochloride, (–)-epinephrine (+)-bitartrate salt, ethidium bromide, histamine
dihydrochloride, ICI-118,551 hydrochloride, indomethacin, (–)-isoproterenol hydrochloride,
dl-normetanephrine hydrochloride, dl-propranolol hydrochloride, and
prazosin hydrochloride were obtained from Sigma-Aldrich Co. (St. Louis, MO, USA);
(–)-(R)-norepinephrine hydrogen tartrate monohydrate, serotonin
creatinine sulfate, and papaverine hydrochloride were obtained from Wako Pure Chemical
Industries (Osaka, Japan); and prostaglandin F was obtained from Fuji Pharma
Co., Ltd. (Tokyo, Japan). (±)-Bupranolol hydrochloride was kindly donated by Kaken
Pharmaceutical Co., Ltd. (Tokyo, Japan).
Atenolol was dissolved in 0.1 N HCl to produce a stock solution of 10−2 M.
indomethacin was dissolved in 100% ethanol to produce a stock solution of 10−2M. All other drugs were prepared as aqueous solutions and diluted with distilled water.
Drugs were added directly to the organ bath and expressed as the molar concentration (M)
in the bath medium.

Preparation and Storage of Biochemical Compounds

Indomethacin, dl-propranolol hydrochloride, hemicholinium-3 bromide, choline chloride, were purchased from Sigma Chemical (Milan, Italy). Methyl [3H]-choline was obtained from NEN™ Life Science Products, Inc. (Boston, MA), and scintillation cocktail Ultima Gold™ from Perkin Elmer (Waltham, MA). GLY and IND were provided by Novartis Pharma (Basel, CH). Indomethacin was dissolved in absolute ethanol and IND in dimethyl sulfoxide; all other compounds were dissolved in distilled water. Fresh solutions were prepared weekly and stored at 4 °C to be used within 7 days.

Preparation of Neurotransmitter Compounds

Preparation and Use of PAF in Research

The following drugs were used in this study: PAF C-16 and apafant (Cayman Chemical, Ann Arbor, MI, USA); TTX, suramin sodium and anti-foam (FUJIFILM Wako Pure Chemical Co., Osaka, Japan.); phentolamine mesylate (Novartis Pharma KK, Tokyo, Japan); dl-propranolol hydrochloride, atropine sulfate salt monohydrate, and indomethacin (Sigma-Aldrich Co., St. Louis, MO, USA); and BSA (fatty acid free, pH 7.0; Nacalai Tesque Inc., Kyoto, Japan).
PAF was dissolved in ethanol (EtOH) to prepare a stock solution of 2 × 10−3 M and stored at −80°C. When using PAF, the EtOH solvent was evaporated, and PAF was redissolved and diluted with 0.25% BSA to prepare solutions of 2 × 10−4–2 × 10−7 M. indomethacin was dissolved in EtOH to prepare a stock solution of 10−2 M. All other drugs were dissolved and diluted with distilled water.

Pharmacological Manipulation of Fear Memory

Midazolam (GobbiNovag S.A.) was diluted in sterile saline (SAL, 0.9% w/v) to a concentration of 3mg/ml and given intraperitoneally (i.p; Bustos et al., 2010 (link)).
DL-Propranolol hydrochloride and D-cycloserine (Sigma-Aldrich) were both dissolved in SAL at concentrations of 10mg/ml and 5mg/ml, respectively, for i.p. injection (Debiec and LeDoux, 2004 (link); Bertotto et al., 2006 (link); Muravieva et al., 2010 (link)). The total volume of drug or an equivalent amount of SAL was 1.0ml/kg in all cases.
For intra-BLA infusions, PROP was dissolved in SAL to a final dose of 5 ug/ul and the amount infused was 1.25 ug/side (Debiec and LeDoux, 2004 (link)).
