Overexpression and silencing of AGAP2-AS1 and TFPI2 in GBM cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
To overexpress AGAP2-AS1 or TFPI2, the full length cDNA sequences of AGAP2-AS1 or TFPI2 were amplified and subcloned into pcDNA3.1 vector (Invitrogen) according to the manufacture’s guidelines. To suppress gene expression, small interfering RNAs (siRNAs) including si-AGAP2-AS1, si-EZH2, si-LSD1 and si-TFPI2 were purchased from RiboBio (Guangzhou, China). GBM cell were seeded into six-well plates and transfected with plasmids, siRNAs or corresponding negative control (Vector or si-NC) using Lipofectamine 2000 (Invitrogen).

Check if the same lab product or an alternative is used in the 5 most similar protocols
LINC00662 overexpression plasmids (LINC00662; 50 ng), empty vector (vector; 50 ng), miR-144-3p mimics (miR-144-3p, 5′-UUCUCGAACGUGUCACGUUUU-3′; 50 nM), mimics negative control (miR-control, 5′-UCAUGUAGUAGAUAUGACAU-3′; 50 nM), siRNAs targeting EZH2 (si-EZH2, 5′-GAGGGAAAGUGUAUGAUAATT-3′; 100 nM), and siRNAs negative control (si-NC, 5′-GGGAAAGAGUAUAUAGUGATT-3′; 100 nM) were purchased from RiboBio Co., Ltd. (Guangzhou, China). SCC-9 and ISG15 cells were transfected with the oligonucleotide or plasmid employing Lipofectamine 2000 transfection reagent (Invitrogen) according to the protocols provided by the manufacturer.

Transfection of miR-506-3p in Colon Cancer Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cell transfection was performed as previously described [28 (link)]. GenePharma has synthesized miR-506-3p-mimic (5ʹUAAGGCACCCUUCUGAGUAGA-3ʹ), miR-506-3p-inhibitor (5ʹ-UCUACUCAGAAGGGUGCCUUA-3ʹ) and corresponding miR-NC (China), si-NC corresponding to small interfering RNA that targets EZH2 (si-EZH2) purchased from RiboBio Company (China). Based on the kit instructions, these reagents were transfected into HT-29 and HCT-8 cells with Liposome 3000 reagent (Invitrogen), the collected cells for subsequent experiments 72 h after transfection.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
