Lentiviral Gene Silencing in BM-MSCs

A lentiviral system obtained from Taiwan National RNAi Core Facility was used for gene silencing in BM-MSCs [28] (link). Lentivirus-infected cells (5×104) were cultured in fresh growth medium (IMDM supplemented with 10% FBS, 1% l-glutamine, 0.1% penicillin–streptomycin, and 10 ng/ml bFGF) in a 6-cm tissue culture plate for 3 days. Culture supernatants were collected and assayed using the RayBio human chemokine antibody array 1 and the RayBio human inflammatory marker antibody array 3 (RayBiotech, Norcross, GA, USA). The assays were performed in accordance with the manufacturer's instructions.
