Bmi1gfp

Manufactured by Jackson ImmunoResearch

Bmi1GFP is a fluorescent protein-tagged fusion protein that can be used to detect and visualize the expression of the Bmi1 gene. Bmi1 is a key regulator involved in various cellular processes such as cell proliferation and stem cell maintenance. The GFP (Green Fluorescent Protein) tag allows for the localization and tracking of Bmi1 expression within cells.

Transgenic mice were Bmi1CreERT/+ (Ref. 21 (link)), Bmi1GFP/+ (Ref. 22 (link)), Rosa26DTA/+ (Ref. 23 (link)), Rosa26YFP/+, Rosa26Tomato/+ (Ai9) and β-actinGFP/+ (all from Jackson Laboratory), all on the C57BL/6 background. The full description of transgenic mouse lines and their application in this study can be consulted in Table I in the online-only Data Supplement. Tamoxifen (Tx) was dissolved in corn oil (Sigma); mice received Tx (i.p.) every 24 h for three consecutive days (225 μg/g body weight daily), or once (7.5 μg/g body weight) for low Tx induction. Since preliminary analysis showed no differences between male and female mice, all experiments were carried out in males and females, as recommended by the US National Institutes of Health.24 (link) Animal studies were approved by the CNB-CSIC Ethics Committee and by the Division of Animal Protection of the Comunidad de Madrid (PA 56/11, PROEX 048/16). All animal procedures conformed to EU Directive 2010/63EU and Recommendation 2007/526/EC regarding the protection of animals used for experimental and other scientific purposes, enforced in Spanish law under Real Decreto 1201/2005.

Transgenic mice used in this study were Bmi1CreERT/+, Bmi1GFP/+, Rosa26mT.mG/+, Rosa26Tomato/+ (All from The Jackson Laboratory), Gli1CreERT/+ [10 (link)], G6PDTg [34 (link)] and Sod3−/− [35 (link)], and all were on the C57BL/6 background. Tamoxifen (Tx; Sigma, T5648) was dissolved in corn oil (Sigma, C8267) and intraperitoneally (i.p.) injected (103 μg/g body weight). Experiments were carried out in male and female mice as recommended by the US National Institutes of Health since preliminary analysis revealed no differences between males and females [36 (link)]. 7-8-week-old mice were used as adult mice unless otherwise indicated in the text. Animal studies were approved by the CNB-CSIC ethics committee and by the Division of Animal Protection of the Comunidad de Madrid (PA 56/11, PROEX 048/16). All animal procedures conformed to EU Directive 2010/63EU and Recommendation 2007/526/EC regarding the protection of animals used for experimental and other scientific purposes, enforced in Spanish law under Real Decreto 1201/2005.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
