Protein Extraction and Western Blot Analysis

Total protein was extracted using RIPA buffer (25 mM Tris-HCl pH 7.6, 150 mM NaCl, 1% NP-40, 1% sodium deoxycholate, 0.1% SDS) supplemented with protease and phosphatase inhibitors (ThermoFisher). Disruption and homogenization of tumor samples was achieved by the use of a TissueLyser II (Qiagen). Protein concentration was determined by Pierce™ BCA Protein Assay Kit (ThermoFisher).
For Western blot analysis, 25 µg of protein per sample was separated by SDS-PAGE and transferred onto a nitrocellulose membrane by semidry blot. For detection of proteins, following antibodies were used: CREB1 antibody 48H2 (CST), Argonaute-2 antibody C34C6 (CST), Maltose Binding Protein antibody ab9084 (Abcam), and β-actin antibody ab8227 (Abcam). As secondary antibody, a horseradish peroxidase conjugated goat α-mouse/rabbit antibody (CST) was used. Chemiluminescent blots were imaged by LAS-3000 Imaging System (Fuji). For densitometric analysis of western blot signal intensity ImageJ software51 (link) was used.

Proteins were separated by sodium dodecyl sulfate polyacrylamide gel electrophoresis and transferred to a polyvinylidene difluoride membrane (Millipore, Billerica, MA, USA). The membranes were blocked with 5% skim milk in the buffer (10 mM Tris [pH 7.5] and 50 mM NaCl) and incubated with primary antibodies overnight at 4°C. The membrane was further treated with secondary antibodies (1:3,000). Immunoreactive bands were visualized by enhanced chemiluminescence system (Amersham, Arlington Heights, IL, USA).
