Quantification of TNF-α Release by hMSCs on Scaffolds

The hMSCs were seeded at a density of 3 × 104 cells/cm2 on scaffolds and on the PLA sample (CTR). At 3 and 7 days, cell supernatants were isolated, centrifuged at 1200 rpm for 5 min and kept at −80 °C until use. ELISA development kit (PeproTech® EC Ltd., London, UK) was used for quantification of Tumor Necrosis Factor-α (TNF-α) released by the cells grown on the scaffolds. Interleukin binding was identified with biotin-avidin detection step, after chromogen 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid (Sigma-Aldrich®, Dorset, UK) treatment with ELISA reader at 405 nm.
