Mitochondrial Function Assay of Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
The mitochondrial functionality was analyzed by HCS Mitochondrial Health Kit (Thermo Fisher Scientific), following the manufacturer’s procedures. Briefly, hFM-MSCs and hAFSCs were treated with 0.1 μM of BPA, BPS, PFOS, and PFOA, alone or in combination, for 24 h. Then, cells were incubated with the MitoStain for 30 min at 37 °C and then fixed in paraformaldehyde 4% for 10 min. Nuclei were counterstained by Hoechst 33342. To quantify the fluorescence, 4 different fields for each sample were acquired by EVOS M7000 (Thermo Fisher Scientific) and analyzed by Celleste Image Analysis Software (Thermo Fisher Scientific).

Jackfruit Bioavailability and Oxidative Stress

Check if the same lab product or an alternative is used in the 5 most similar protocols
Jackfruits were obtained from Hainan Province, China. Acrylamide was purchased from Aladin. Pepsin (2000 U/mL), pancreatin (800 U/mL), bile, dulbecco’s modified eagle medium (DMEM) culture media, and Folin–Ciocalteu’s phenol reagent were purchased from Sigma Chemical (St Louis, MO, USA). Caco-2 was obtained from the Shanghai Institute of Cell Biology at the Chinese Academy of Sciences (Hangzhou, China). Hoechst 33342 and PI were purchased from Beyotime Institute of Biotechnology (Shanghai, China). HCS Mitochondrial Health Kit and Cell ROX® Oxidative Stress Reagent were purchased by Thermo Scientific (San Jose, CA, USA). All other reagents were of analytical grade.

Simultaneous Mitochondrial and Cytotoxicity Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
The HCS Mitochondrial Health Kit (H10295, Thermo Fisher Scientific) was used to measure mitotoxicity and cytotoxicity simultaneously in the same cell. Briefly, cell staining solution was prepared by adding 10.5 μl of the MitoHealth stain solution and 2.1 μl of Image-iT® DEAD Green™ viability stain to 6 ml complete medium according to the manufacturer’s protocol. The neuronal cultures were incubated with the cell staining solution for 30 min under culture conditions, followed by adding the counterstain/fixation solution for 15 min at room temperature. The red MitoHealth stain corresponding to mitochondrial membrane potential (MMP) and DEAD Green (cytotoxicity) were captured under a LSM700 Meta confocal laser microscope (Carl Zeiss).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
