Everolimus Treatment of MCL Cell Line

Granta 519 cells were obtained from the American Type Culture Collection (Manassas, Virginia). This cell line has been previously well characterized as a model of MCL. Cells were tested for mycoplasma contamination using the MycoAlert mycoplasma detection assay kit from Lonza (Basel, Switzerland) and found to be negative. The cell line was verified to be authentic by the University of Arizona Genomics Core. Cells were routinely grown in Dulbecco modified Eagle medium (DMEM) supplemented with 10% fetal bovine serum (HyClone Thermo Scientific, Waltham, Massachusetts) in a humidified incubator at 37°C and 5% CO2. Per test parameters, the DMEM either had 4.5 or 1.0 g/L of glucose. Cells were treated with 10 nmol/L everolimus dissolved in biological grade 100% dimethyl sulfoxide (DMSO).

Cell Line Maintenance and Validation

GL261 cells were obtained from NCI Division of Cancer Treatment and Diagnosis Tumor Repository. Human Jurkat T cells (clone E6–1) were obtained from ATCC. Cells were maintained in liquid nitrogen in Bambanker cryopreservation media until further use. Cells were tested for Mycoplasma using Lonza MycoAlert Mycoplasma Detection Assay Kit prior to any use. Cells were used within 3–5 passages.
