Electrospinning of Polyester Biomaterials

Three types of polyesters were used: PCL (Sigma-Aldrich, USA, , , Tg = −60 °C), PLCL 70 : 30 (Resomer® LC703S, Evonik, Germany, , , inherent viscosity = 1.3–1.8 dl g−1, Tg = 32–42 °C) and PLLA (PL49, Corbion, Netherlands, , , inherent viscosity = 4.9 dl g−1, Tg = 58 °C). Solvents used in electrospinning process—acetic acid (AA) (purity degree 99.5%), formic acid (FA) (purity degree ≥ 98%), and hexafluoroisopropanol (HFIP) (purity degree 98.5%) were purchased from Poch (Poland), Sigma-Aldrich (USA), and Iris Biotech GmbH (Germany), respectively. Ethylenediamine (EDA) (purity degree 99.5%), isopropanol (purity degree 99.8%), ninhydrin (purity degree 99%), ethanol (purity degree 99.8%), glutaraldehyde (conc. 25%) and tetrahydrofuran (THF) (for HPLC) were purchased from Chempur (Poland). Chloroform (LiChrosolv®, for liquid chromatography) was procured from Sigma-Aldrich (USA). For all polymers, and were determined experimentally.

Synthesis of Functional Polymers via ATRP

The methyl methacrylate (MMA; Alfa Aesar, Warsaw, Poland) was dried using molecular sieves (type 4Å, Chempur, Piekary Śląskie, Poland). The [2-(methacryloyloxy)ethyl]-trimethylammonium chloride (TMAMA/Cl; 80% aq. solution, Sigma-Aldrich, Poznań, Poland) was dried to a constant weight under reduced pressure. Methanol and tetrahydrofuran (THF) were purchased from Chempur (Piekary Śląskie, Poland) and dried over the same molecular sieves as MMA. Copper(I) bromide (CuBr; Fluka, 98%, Steinheim, Germany) was purified using a procedure described previously in the literature [42 (link)]. N,N,N′,N,N″-pentamethyldiethylenetriamine (PMDETA, 98%), ethyl 2-bromoisobutyrate (EBiB, 98%), potassium clavulanate (KCLV), and bis(trifluoromethane)sulfonimide lithium salt (LiTf2N) were obtained from Sigma Aldrich (Poznań, Polska). Sodium p-aminosalicylate (NaPAS, 98%), sodium piperacillin (NaPIP, 99%), and sodium fusidate (NaFUS, 98.8%) were purchased from Alfa Aesar (Warsaw, Poland) and used without further purification.

Synthesis of Thermoresponsive Polymer Brushes

Tetraethoxysilane (TEOS, 98% GC), hexadecyltrimethylammonium bromide (CTAB, 98%), α-bromoisobutyryl bromide (BIBB, 98%), (3-aminopropyl)triethoxysilane (APTES, >98%), copper (I) bromide, N-isopropylacrylamide (NIPAM, 97%), hydrochloric acid (37%) were purchased from Sigma Aldrich (Darmstadt, Germany). Triethylamine (TEA, >99%), N,N,N′,N″,N″-pentamethyldiethylenetriamine (PMDETA, >99%) were purchased from TCI (Tokyo, Japan). Rhodamine 6G (Rh6G, 99%) was purchased from Acros Organics (Geel, Belgium). Ammonia solution (30%, p.a.), ethanol (96%, p.a.), tetrahydrofuran (THF, p.a.) were purchased from Chempur (Piekary Śląskie, Poland). THF was dried under molecular sieves prior to use. N-isopropylacrylamide was purified by recrystallization from n-hexane. All the other chemicals were used as received. Deionized water was used in all the procedures.

Biofunctionalized PLCL Scaffold Fabrication

Hexafluoroisopropanol (HFIP) (purity degree 98.5%) was procured from Iris Biotech GmbH (Marktredwitz, Germany). Ethylenediamine (EDA) (purity degree 99.5%), sodium hydroxide (NaOH), isopropanol (purity degree 99.8%), glutaraldehyde (GTA, conc. 25%), and tetrahydrofuran (THF) were purchased from Chempur (Piekary Śląskie, Poland). Gelatin (G1890, type A, gel strength ~300 g Bloom) and bicinchoninic acid (BCA) assay kit (QuantiPro™, for 0.5–30 µg/mL protein conc.) was supplied by Sigma-Aldrich (St Louis, MO, USA). N-hydroxysuccinimide (NHS), 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride (EDC), Dulbecco’s Modified Eagle Medium (DMEM), Phosphate-Buffered Saline (PBS) pH 7.4, Foetal Bovine Serum (FBS), Trypsin EDTA, antibiotic (Penicillin-Streptomycin 10,000 U/mL), Presto Blue, NucBlue, and ActinGreen were purchased from Thermo Fisher Scientific (Waltham, MA, USA). Mouse fibroblast L929 were purchased from Sigma-Aldrich (ATTC).
