Deethylametryn (99.9% w/w) and didealkylametryn (98.7% w/w) were purchased from TLC PharmaChem, Inc. (Ontario, Canada). Prometryn sulfoxide (98.0% w/w) and prometryn sulfone (96.0% w/w) were customized and synthesized by Shanghai Nafu Biotechnology Co., Ltd. (Shanghai, China). Acetonitrile and methanol were purchased from Fisher Scientific (Fair Lawn, NJ, USA). Formic acid (optima LC/MS) was supplied by Fisher Scientific (Shanghai, China). Ultrapure water was obtained from Sartorius Lab Instruments GmbH & Co. KG (Göttingen, Germany). All chemicals were of analytical reagent grade and were used without further purification. Modified Hoagland nutrient solution with 16 essential elements and N, P, K, Ca, and Mg concentrations of 115, 23, 185, 80, and 24 mg/L, respectively, was purchased from Fushan Yigeng Agricultural Technology (Yantai, Shandong, China).

Reagent Procurement for Research Protocols

All chemicals were purchased from Sigma-Aldrich unless otherwise specified. Tetrahydrofuran (HPLC grade) (THF), ethanol, absolute (HPLC grade) (EtOH), ethyl acetate (analytical grade reagent), and Formic Acid Optima LCMS were purchased from Fisher Scientific. Acetonitrile and methanol (MeOH) were obtained from VWR Chemicals. Yeast extract was obtained from Fluka. Bacto™ Tryptone was obtained from BD Biosciences. Luria Bertani (LB) agar was obtained from Carl Roth. Gibco® HBSS (1x) Hanks' Balanced Salt Solution and Gibco® PBS was obtained from Life Technologies. Purified water was prepared by a Milli-Q water purification system (Merck Millipore, Billerica, MA, USA) (called water in the manuscript).

Biotoxin Standards Preparation and Use

All acetonitrile (ACN), methanol (MeOH), isopropyl alcohol (IPA), and acetone (ACT) used were LC-MS grade (Honeywell Burdick & Jackson, MI, USA). Ultrapure water was prepared using a Milli-Q system (Millipore, Billerica, MA, USA). Formic Acid Optima LC/MS (Thermo Fisher Scientific, Waltham, MA, USA), Ammonium Formate Optima LC/MS (Thermo Fisher Scientific, Waltham, MA, USA), and ammonium hydroxide eluent additive for LC-MS (≥25%, Honeywell Fluka, Charlotte, NC, USA) were used for mobile phases. Formic Acid 98% (EMD Millipore, Merck, Germany) was used for the carryover wash solution.
The following biotoxin certified reference materials were purchased from the National Research Council Canada: azaspiracid-1 (CRM-AZA1-b), azaspiracid-2 (CRM-AZA2-b), dinophysistoxin-1 (CRM-DTX1-b), dinophysistoxin-2 (CRM-DTX2-b), okadaic acid (CRM-OA-d), pectenotoxin-2, (CRM-PTX2-b), and yessotoxin (CRM-YTX-c). Brevetoxin-2 was purchased from Abcam (ab143469). A microcystin-RR, -YR, -LR mixed solution was purchased from Sigma Aldrich (33578-1ML). Karlotoxin 3 (KmTx 3) was purified from Karlodinium veneficum by Allen Place (UMCES, Maryland). Goniodomin A was purified from Alexandrium monilatum by Drs. Constance and Tom Harris [37 (link)] and provided by Dr. Kimberly Reece (VIMS).

Deer Antler Peptide Biomarker Discovery

Trypsin (proteomics grade), guanidine hydrochloride, hydroxymethyl aminomethane (Tris), ethylenediaminetetraacetic acid (EDTA), ammonium bicarbonate, and acetic acid (biotech grade) were purchased from Sigma-Aldrich (St. Louis, MO, USA), formic acid (optima LCMS) was obtained from Thermo Fisher Scientific (Waltham, MA, USA), acetonitrile (gradient grade) and methanol (gradient grade) was purchased from Merck KGaA (Darmstadt, Germany). Ultrafiltration centrifuge tubes (3 kDa, 0.5 mL) were purchased from Merck Millipore (Burlington, MA, USA). Water was prepared on a Milli-Q A 10 Gradient system by Millipore (Schwalbach, Germany).
Twenty-three batches of deer antlers were collected from markets in different regions of China. The species of deer antlers were identified by the Shandong Academy of Agricultural Sciences using the DNA barcode method; 3 batches of reindeer antlers were identified by the method provided by literature [3 (link)], and the sample information is shown in Table S1. Seven species of deer antlers were used to discover peptide biomarkers, and all samples were used for validation. The peptide biomarkers shown in Table 1 were chemically synthesized by GL Biochem (Shanghai, China), Yuan Peptide (Nanjing, China) and China peptides (Suzhou, China).

LC-MS grade water was produced by Milli-Q Merck Millepore (Massachusetts, USA) water system using LC-pak Polisher cartridge containing C18 reverse phase silica.
