Tris-HCl, PBS, aniline, iodoacetamide, dithiothreitol (DTT), urea, protease inhibitor cocktail solution, sodium(meta)periodate, Triton X-100, SDS, NH4HCO3, and glycerol were purchased from Sigma-Aldrch (St. Louis, MO). Pierce C18 spin columns were purchased from Glygen Corp (Columbia, MD). Fluorescently labeled monoclonal antibodies to murine Gr1 (clone RB6-8C5), CD11b (clone M170), CD47 (clone miap301), and isotype-matched controls were from BioLegend (San Diego, CA). Deionized water used for all experiments was obtained from a Milli-Q A10 system.

Proteomic Analysis of Modified Proteins

Carnitine Quantification by LC-MS

Quantification of Deuterium-Labeled Ibrutinib and Metabolites

Deuterium‐labeled 4β‐HC ([2H7]4β‐HC, d7‐4β‐HC, H917982) and deuterium‐labeled cholesterol ([2H7]cholesterol, d7‐cholesterol, C432503) were purchased from Toronto Research Chemicals. Dimethyl sulfoxide (DMSO), Optima liquid chromatography mass spectrometry LC–MS grade water, Optima LC–MS grade acetonitrile, Optima LC–MS grade methanol were obtained from Fisher Scientific. All other chemicals and reagents were of analytical grade or higher.
As described previously,23 an NADPH‐regenerating system, consisting of solution A (26 mM NADP+, 66 mM glucose 6‐phosphate, 66 mM MgCl2 in water) and solution B (40 U/ml glucose 6‐phosphate dehydrogenase in 5 mM sodium citrate), was purchased from Corning Life Sciences. InVitroGRO Krebs–Henseleit buffer (KHB) medium (product no. Z99074) and thawing (HT) medium (product no. 990006) were purchased from BioIVT.

Analytical Protocols for Food Ingredients

Quantification of Cyanotoxins in Aquatic Samples

Lyophilized analytical standards of microcystin-LA, LF, LR, LW, LY, RR, YR, and nodularin, all with manufacturer reported purities of ≥95%, were purchased from Enzo Life Sciences, Inc. (Framingdale, New York, USA). Stock solutions were prepared at concentrations of 10 μg/mL in methanol. Certified reference materials (CRMs) of microcystin-LR, RR, and nodularin, in methanol, as well as anatoxin-a, cylindrospermopsin, and saxitoxin were purchased from the National Research Council Canada (Ottawa, Ontario, Canada). All MC standards were stored in the dark at −20 °C. All other standards were stored according to the manufacturer's recommendations. For LC-MS/MS and PPIA analyses, Optima LC/MS Grade acetonitrile, methanol, and water were purchased from Fisher Scientific (Pittsburgh, PA, USA) and used for sample preparation and LC eluents. Trifluoroacetic acid (Chromasolv® for HPLC, >99.0%) and Tween 20 for PPIA extractions were purchased from Sigma Aldrich (St. Louis, MO, USA).
