RPE1 cells survival assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
RPE1 cells lines were seeded at 3000 cells/well in 6-well plates and allowed to attach overnight. TULIP2 constructs were induced with 1 µg/mL doxycycline prior treatment. Olaparib (Bio-connect) was added at different concentrations for 24 h. After treatment, medium was refreshed. Hydroxyurea (Sigma) was added at different concentrations and medium was refreshed after 16 h treatment. Subsequently, cells were allowed to grow for 10 days and fixed for 20 min in 4% paraformaldehyde (PFA) in PBS. Cells were stained with Crystal Violet 0.05% for 30 min and washed with water. Afterward, Crystal Violet was re-solubilized in methanol and O.D.595 was measured in the VICTOR X3 Multilabel Plate Reader 2030-0030 (Perkin Elmer). GraphPad Prism 10 was used for statistical analysis and the value of untreated cells was set at 100% survival.

Regorafenib (RGF), regorafenib M2 (RGF M2), olaparib (OPR), vemurafenib (VMF), cobimetinib (CBT), niraparib (NPR), cabozantinib (CBZ) and dabrafenib (DBF) were obtained from Bio‐Connect BV (Huissen, The Netherlands). The isotopes 13C2H3‐regorafenib, 2H8olaparib, 13C6‐vemurafenib, 13C6‐cobimetinib, 13C6‐niraparib, 2H4‐cabozantinib and 2H9‐dabarefenib used as internal standards, were acquired from Alsachim (Illkirch, France). Dimethyl‐sulfoxide (DMSO, Seccosolv) and acetonitrile (ACN, Lichrosolv) were purchased from Merck (Darmstadt, Germany). Formic acid was obtained from Sigma‐Aldrich (Zwijndrecht, The Netherlands). High‐purity Milli‐Q water was produced using a MilliQ Gradient water purification system (Millipore, Amsterdam, The Netherlands). Ethylenediaminetetraacetic acid (EDTA) plasma was prepared from EDTA whole blood obtained from Sanquin (Amsterdam, The Netherlands).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
