Azoxymethane (AOM) was procured from Sigma-Aldrich Co. (St. Louis, MO, USA). Sodium carboxymethylcellulose (CMC-Na) was purchased from Sangon Biotech (Shanghai, China). Lipopolysaccharide (LPS) was obtained from Sigma-Aldrich Co. (St. Louis, MO, USA). Antibodiesfor CEA, Ck8, Ck18, p53, p-NF-κB-p65, T-NF-κB-p65, p-state-3, T-state-3, and β-actin were purchased from Affinity Bioreagents (Colorado, USA). BCA assay kits were obtained from Thermo (USA). The STAT3 inhibitor HO-3867 was purchased from Selleck Chemicals (Houston, TX, USA). ELISA kits were purchased from Tainuo (Shanghai, China).

S-Benzylisothiourea (cat no: S9138) was ordered from Sigma-Aldrich (USA). The primary antibodies listed below were ordered from Cellular Signaling Technology (USA): caspase-3 (cat no:9662), Mcl-1 (cat no:94296), Bcl-2 (cat no:15071), Bcl-xL (cat no:2762), Bak (cat no:6947), Bax (cat no:94296), and DMT1 (cat no:15083). The following primary antibodies listed were procured from Abcam (USA): FPN1 (cat no: ab239583), TFR (cat no: ab214039), FTL (cat no: ab75973), FTH (cat no: ab183781), and p53 (cat no: ab26). The secondary antibody was procured from Abclonal (China). Sigma-Aldrich supplied all other routine chemicals.

Pharmacological compounds used in this study included recombinant human IL-6 (Preprotech, London, UK), recombinant human IL-6R (Preprotech), rabbit monoclonal antibody against IL-6R (R&D Systems, Inc. Minneapolis, MN, USA), and goat polyclonal antibody against TRPM7 (Santa Cruz Biotechnology, Santa Cruz, CA, USA). The following compounds were purchased from Sigma−Aldrich (St. Louis, MO, USA): the JAK2 inhibitor AG490, the PI3K inhibitor wortmannin, the PLC inhibitor U73122, TEACL, TTX, nimodipine, CsCl, choline chloride, spermine, GdCl3, and 2-APB. The STAT3 inhibitor HO-3867 and the Erk inhibitor PD98059 were purchased from Selleck Chemicals (Houston, TX, USA). Dimethyl sulfoxide (DMSO) was used to dissolve hydrophobic inhibitors, including 2-APB, AG490, PD98059, HO-3867, and U73122, with resulting final concentrations of DMSO of less than 0.1% (v/v).
