Anticoagulant Reversal and Platelet Aggregation

Check if the same lab product or an alternative is used in the 5 most similar protocols
Thirty-three male Wistar rats weighing 182–212 g were randomly assigned to 3 groups, anesthetized by an intraperitoneal injection of pentobarbital (45 mg kg−1), and placed in a supine position on a heated operating table. Rats were intravenously injected into the right femoral vein with HBC (4.5 mg kg−1) 5 min after injection with HS (6 mg kg−1). Twenty minutes after HS administration, the blood samples were collected from the heart and drawn into 3.13% trisodium citrate. Neutralization of HS by HBC was monitored by measuring aPTT, PT, anti-fXa and anti-fIIa activity, and platelet aggregation. Coagulation parameters were determined as described above whereas platelet aggregation was measured after incubation of sodium citrate-anticoagulated blood with 0.9% NaCl solution (volume ratio 1 : 1) for 20 min at 25 °C and then for 15 min at 37 °C. The changes in impedance registered as aggregation curve were recorded for 6 min after collagen addition (7.5 μg mL−1) using a Chrono-log aggregometer (Chrono-log Corp.). Four parameters were automatically calculated: maximal extension (MaxA), lag phase (Lag), the slope of platelet aggregation (Slp), and area under the curve (AUC). The white blood cell count (WBC) was automatically performed using an animal blood counter (scil Vet abc Plus, Horiba, France).

Longitudinal Blood Cell Monitoring

Check if the same lab product or an alternative is used in the 5 most similar protocols
Eight to ten mice of each three genotypes (WT, HET and KI) were selected for monitoring over the course of a year. Blood sampling was performed under isoflurane anesthesia every two months via venous puncture. Blood counts were determined with an automatic cell counter (Scil Vet ABC Plus, Horiba Medical, France). Mice were kept for 14–16 months.

Automated Coagulation and Hematology Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
The activated partial thromboplastin time (APTT), prothrombin time (PT), also expressed as the QUICK index, and INR and fibrinogen concentration were determined automatically using the Coag-Chrom 3003 apparatus (Bio-ksel, Poland) and standard dedicated laboratory reagents (Bio-ksel, Poland). Blood count was performed using a ScilVet ABC Plus+ hematological analyzer (HORIBA ABX, France). The counting of blood cells was based on the volumetric impedance method.

Type 2B vWD Mouse Model Characterization

Check if the same lab product or an alternative is used in the 5 most similar protocols
The type 2B vWD knock-in mouse model (p.V1316M) has been described elsewhere.11 (link) Mice homozygous for the p.V1316M mutation are referred to hereafter as “2B mice”, and their control litter-mates are referred as wild-type (WT) mice. Platelet counts were determined with an automated analyzer (Scil Vet ABC Plus, Horiba Medical). Male and female mice were used indifferently. The study was approved by the local animal care and use committee (reference: APAFIS#1294-2015072816482568).

Blood Morphology and Coagulation Factors

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood morphology test was performed using a hematological analyzer (ScilVet ABC Plus+, HORIBA ABX, Grabels, France). Plasma levels of tissue factor (TF), tissue plasminogen activator (t-PA), and plasminogen activator inhibitor type 1 (PAI-1) were measured by enzyme immunoassays (Rabbit Monoclonal Antibody Anti Rat TF; ImmunoKontact AMS Biotechnology, Abingdon, UK; Rat Active tPA ELISA Kit, Innovative Research, Novi, MI, USA; Rat Active PAI-1 ELISA Kit; Innovative Research, Novi, MI, USA, respectively).

Plasma Coagulation Factors Determination

Check if the same lab product or an alternative is used in the 5 most similar protocols
Tissue factor, t-PA and PAI-1 plasma levels were determined by enzyme immunoassay (Rat Tissue Factor ELISA Kit, MyBioSource, Inc., San Diego, CA, USA; Rat Active t-PA ELISA Kit and Rat Active PAI-1 ELISA Kit, Innovative Research, Inc., Novi, MI, USA) in a microplate reader (ELx808, BioTek Instruments, Inc., Winooski, VT, USA) according to the manufacturer’s instructions. A blood morphology test was carried out using the hematological analyzer ScilVet ABC Plus+ (HORIBA ABX, Montpellier, France). A volumetric impedance method of counting blood cells was applied [54 (link)] for determination of white blood cells (WBCs), red blood cells (RBCs), medium cell volume (MCV), medium content of hemoglobin (MCH), medium cell hemoglobin concentration (MCHC), hemoglobin (HGB), platelet count (PLT), as well as automatic calculation of hematocrit (HCT).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
