DNA fragmentation (DNA fragmentation kit-TaKaRa; 6137) was detected by agarose gel electrophoresis. The cell suspension remaining after Trypan blue (970 µL, 2 × 106/mL) was lysed in 5 mM Tris, 20 mM EDTA, and 0.5% Triton X-100 (pH 8.0) at 4°C for 1 hour and centrifuged at 13,000 rpm for 15 minutes. DNA was ethanol precipitated overnight at -20°C, further treated with proteinase K and RNAse, loaded on 1.0% agarose gels, and electrophoresis maintained at a constant voltage of 70 V for 2 hours. Fragmentation of DNA was visualized with ethidium bromide using the Bio-Rad gel doc.
