Buccal Cell-Derived Genomic DNA Extraction

Genomic DNA (gDNA) from all oocyte donors was obtained using buccal cell swabs (Isohelix, Cell Projects Ltd). Extraction of the gDNA from the swabs was performed using a proteinase K extraction kit to a final volume of 30 μl, following the manufacturer’s instructions. DNA from all three products of meiosis was obtained by lysis of the cells and whole-genome amplication (WGA). The PCR tubes containing the samples were brought to an end volume of 4 μl with PBS and REPLI-g Single Cell Kit multiple displacement amplification (SureMDA, Illumina) or PCR library based SurePlex amplification (Illumina) was performed according to the manufacturer’s instructions. MDA was performed with a short 2h incubation.
