Quantifying HUVEC Tube Formation in Matrigel

Matrigel tube formation assay was used to determine HUVECs tube formation capacity. Matrigel (Corning, USA) was thawed at 4°C overnight. The liquid Matrigel was spread into 96-well plates (50 μL per well) and incubated in a 5% CO2 incubator at 37°C for 40 minutes. Cell suspensions were counted using a Cellometer-Mini Automatic Cell Counter (Nexcelom Bioscience, Lawrence, MA, USA). HUVECs (5 × 103 per well) were seeded on the Matrigel-coated plates and incubated for 6 hours. The network formation in Matrigel was observed using an inverted light microscope. The Image J software plugin “Angiogenesis Analyzer” quantified the extent of tube formation before statistical analysis.

Cell Dissociation and Counting
