Western Blot Analysis of Cellular Proteins

Cells and tumor tissues were washed with ice-cold PBS and lysed with RIPA buffer containing protease inhibitors. Proteins were quantified using a BCA kit after centrifugation at 4 °C, separated by SDS-PAGE, and transferred to polyvinylidene difluoride membranes (pore size, 0.22 μm, Millipore, USA). Membranes were blocked with 5% skim milk and incubated with primary antibodies overnight at 4°C. The antibodies used were as follows: RACGAP1 (Proteintech, cat no. 13739-1-AP and Abcam, cat. no. ab97315), GAPDH (Proteintech, cat no. 60004-1-Ig and Abcam, cat. no. ab8245); MCM3 (Proteintech, cat no. 15597-1-AP); YY1 (Proteintech, cat no. 66,281–1-Ig), goat anti-mouse IgG (H + L)/HRP conjugate (Proteintech, cat no. SA00001-1), and goat anti-rabbit IgG (H + L)/HRP conjugate(Proteintech, cat. no. SA00001–2). Chemiluminescent imaging was performed using the Tanon 5200 system in conjunction with the SuperSignal West Pico Plus reagent kit (Thermo Fisher Scientific, Waltham, MA, USA), and the target proteins were analyzed by ImageJ software.
