Valerian-Hops Extract Inhibits Acetic Acid-Induced Writhing in Mice

As described before [24 ], in this experiment five groups of mice were treated intraperitoneally (i.p) as follows: the first group received normal saline as vehicle control; group II received 10 mg/kg diclofenac sodium (Sigma- Aldrich). Valerian-hops extract was given to groups two to five (50, 100, and 200 mg/kg, respectively). Acetic acid (0.6%, purity ≥99.0%, 10 ml/kg) (Sigma- Aldrich) was administered for each mouse 60 min after Valerian -hops extract, vehicle, or diclofenac sodium treatments. Complete writhing was recorded and counted for 30 min after Acetic acid treatment. The occurrence of body elongation, abdomen contraction, pelvis ending twisting, and/or trunk twisting associated with limb extension was counted as writhing. Inhibition of writhing was considered as the results of this experiment which were represented as percent according to the following formula:
Percent of inhibition of writhing (PIW) = [(Writhing number (Control) – writhing number (treatment))/Writhing number (control)] × 100.

Synthesis and Electrochemical Analysis of DF-CMK-3

Synthesis of DF-CMK-3: diclofenac sodium (Sigma-Aldrich, St. Louis, MO, USA) and distilled water (Merck, Darmstadt, Germany) were used during synthesis.
Electrochemical measurements: Appropriate volumes of 1 M CH3COONH4 and 1 M HCl (Sigma-Aldrich, St. Louis, MO, USA) were used to obtain 1 M buffer solution (CH3COONH4, CH3COOH, and NH4Cl) with a pH of 5.6. The standard solutions of Fe3+, Cd2+, Zn2+, Cu2+, Ni2+, Sb3+, Pb2+, NO2, NO3, Cl, and Triton X-100 (Merck, Darmstadt, Germany) were used to study the selectivity of the proposed procedure, and 96% ethanol solution (Merck, Darmstadt, Germany) was used to prepare 10 and 1 mM roxarsone (ROX, AK Scientific, Union City, CA, USA). The ROX solution with a concentration of 0.01 mM was prepared daily in 0.1 M PBS buffer with a pH of 7.2. The wastewater samples were obtained from the Municipal Water Supply & Waste Water Treatment Company Ltd. (Lublin, Poland), while river water samples were collected from the Vistula River (Sandomierz, Poland). The ROX spiked samples were filtered (0.22 µm Millipore filter).

Solvent-Assisted Diclofenac Drug Formulation

In Silico Screening for COX Inhibitors

The Schrodinger Maestro molecular modeling package (Schrodinger LLC, New York, NY, USA) was used in all virtual screening modeling steps. QikProp tools are an accurate, rapid, and simple-to-use method for predicting molecular properties. GraphPad Prism 7 software was purchased from (GraphPad Software, Inc., San Diego, CA, USA). Synthesis and Characterization of NC-8

NC-8 (≥97.0% purity) was synthesized from isosteviol (Fig. 1) as described in our previous report [15 (link)]. The purity and chemical composition were verified by an elemental analysis using an Elementar Vario EL cube for CHNS (Elementar Analysensystem, Hanau, Germany). The internal standard (IS) used was diclofenac sodium (purity, 99%), which was purchased from Sigma (St. Louis, MO, USA). Water was purified by an RDI reverse osmosis/deionizer system (Luton Technic, Taipei, Taiwan). Blank rat plasma was collected in our laboratory from male Sprague–Dawley (SD) rats purchased from BioLASCO Taiwan (Taipei, Taiwan).

Comparative Topical Diclofenac Formulations

The formulations tested (Table 1) were Pennsaid® (diclofenac topical solution 2%) (Mallinckrodt Brand Pharmaceuticals, Inc., Staines-upon-Thames, UK), Voltaren® (diclofenac topical gel 1%) (Novartis, Basel, Switzerland) and Solaraze® (diclofenac topical gel 3%) (PharmaDerm, Princeton, NJ, USA). Methanol, acetonitrile, and water (liquid chromatography-mass spectrometry [LC-MS] grade) were purchased from Fischer Scientific (Fair Lawn, NJ, USA). All other chemicals were of the highest quality available.
