Phenylhydrazine-Induced Anemia in Mice

129/SvImJ (wild-type), 129S4/SvJae (wild-type), or 129S4/SvJae-Pparαtm1Gonz/J female mice (The Jackson Laboratory) 6 – 8 weeks of age, randomized by weight, were pretreated with GW7647 (100 μg/kg) for 3 days (days −3 – −1). On day 0, the mice were injected with phenylhydrazine (PHZ) (60 mg/kg). GW7647 injection was continued during days 0 – 6. Whole blood samples were collected at each day of day 1 – 5, and day 7 and 9 for Complete Blood Count (CBC) analyses.
4 to 6 week old Nan/+ mutant mice (officially designated as Klf1Nan; http://www.informatics.jax.org/allele/MGI:1861107), obtained from the Jackson Laboratory, were injected with GW7647 (100 μg/kg) for 18 days. Mice were randomized by weight. Whole blood samples were collected every 3 days for CBC analyses.
All mouse procedures were approved by the Animal Care and Use Committees of the Massachusetts Institute of Technology (Cambridge, MA).

Body Composition Analysis in Mice

All in vivo experiments were carried out with male A/J,
C57BL/6J, AKR/J, DBA/2J, 129SVImJ and (B6 × DBA/2J)F1 mice purchased
from the Jackson Laboratory (Bar Harbor, ME). Mice were maintained in a
temperature-controlled room (23°C) with a 12-h light/12-hr dark cycle.
The mice were reared under conventional conditions and fed PicoLab Rodent Diet
20 (Lab Diet; 13% kcal fat) until 16 weeks of age. A subset of mice was fed high fat diet (D12331; 58 kcal% fat) for 8 weeks (8–16 weeks of age). Body composition (body fat, lean mass and free fluid) was
analyzed by Minispec NMR (Bruker) which uses the contrasting hydrogen density
and/or hydrogen spin properties from adipose tissue and muscle for estimating
body composition. A quality control check of NMR parameters using a standard
provided by the manufacturer was performed at the beginning of each day of
testing. All animal experiments were approved by the Pennington Biomedical
Research Center and Maine Medical Center Research Institute Institutional Animal
Care and Use Committees and in accordance with National Institutes of Health
guidelines for care and use of laboratory animals.

Genetic Diversity in Mouse Models

The mice used in this study were F1 hybrids of C57Bl/6J reference females with wild-derived strains 129/SvImJ (B6x129), C3H/HeJ (B6xC3H), CAST/EiJ, (B6xCAST), PWK/PhJ (B6xPWK), and SPRET/EiJ (B6xSPRET). 129/SvImJ and C3H/HeJ hybrid females were acquired from the Jackson Laboratory (8 weeks old, housed 4/cage). CAST/EiJ, PWK/PhJ, SPRET/EiJ inbred males were acquired from the Jackson Laboratory and were bred to C57Bl/6J female mice at FHCRC. Mice were maintained on a 12-h light, 12-h dark schedule with lights turned on at 7 a.m. The housing room was maintained at 20–24 °C with 30–70% relative humidity. Mice have housed in individually ventilated cages (Allentown) with 75 square inches of floor space and 60 air changes/hour. Biofresh cage bedding was (Absortion Corp) at 1/8 inch depth and autoclaved on site. Water and Purina 5053 (irradiated) were available ad libitum. Nestlet material (Envigo’s diamond twist 7979C, also irradiated) was present in each cage for enrichment. Autoclavable certified igloos (Bio-serv) were provided in some cages. Mice were housed in a barrier facility that is AAALAC accredited. Mice were sacrificed at 8 weeks of age by CO2 asphyxiation. All work was approved by the Institutional Animal Care and Use Committee (IACUC) of the Fred Hutchinson Cancer Research Center (FHCRC).
