HPLC-MS Analysis of Chemical Compounds

Check if the same lab product or an alternative is used in the 5 most similar protocols
Chromatographic analysis was performed on a Dionex Utimate 3000 Series HPLC system (Thermo Scientific, Shanghai, China) equipped with a quaternary solvent delivery system and a column temperature controller. All the samples were analyzed at a column temperature of 30 °C on a Diamosil-C18 column (4.6 mm × 250 mm, 5 μm, Dema, Beijing, China). The mobile phase consisted of water with 0.1% formic acid solution (eluent B), and acetonitrile (eluent A) using a gradient elution mode of 10% A at 0–3 min, 10–15% A at 3–5 min, 15–26% A at 5–60 min, 26–66% A at 60–110 min, 66% A at 110–120 min. The flow rate was 1.0 mL/min, and the injection volume was 10 μL.
A mass spectrometer (Thermo Scientific, LTQ-Orbitrap XL, Shanghai, China) was applied to MS detection. The operation conditions were as follows: capillary temperature, 350 °C; apci vaporizer temperature, 300 °C; sheath gas flow, 30 L/min; aux gas flow, 10 L/min; source voltage, 3 kV; capillary voltage, −35 V; Tube Lens, −110 V. Mass spectra were recorded across the range of m/z 50–2000. Thermo Workstation Acquisition Software Xcalibur Version 2.2 and UHPLC analysis Software Chromeleon Version 7.0 were utilized for system control, data acquisition and data processing.

Fingerprint analysis and quantitative analysis were conducted on a Dionex Utimate 3000 Series HPLC system (Thermo Fisher, Shanghai, China) equipped with a quaternary solvent delivery system, an ultraviolet detector with full wavelength scanning, and a column temperature controller.
All samples of fingerprint analysis and quantitative analysis were analyzed at a column temperature of 30 °C on a Thermo Scientific-C18 column (4.6 × 250 mm, 5 μm, Thermo Fisher, Shanghai, China). The mobile phase consisted of 0.1% phosphoric acid aqueous solution (eluent A) and acetonitrile (eluent B), the gradient elution mode was set as follows: 2–7% B at 0–6 min, 7–9% B at 6–10 min, 9% B at 10–15 min, 9–13% B at 15–35 min, 13% B at 35–40 min, 13–19% B at 40–54 min, 19–29% B at 54–66 min, 29–80% B at 66–103 min, and 80% B at 103–108 min. The injection volume was 10 μL and the flow rate was 1.0 mL/min.

HPLC-ELSD Analysis of Compound Fingerprints

Check if the same lab product or an alternative is used in the 5 most similar protocols
Fingerprint analysis was performed on Dionex Utimate 3000 Series HPLC system (Thermo Fisher, China) equipped with detector of ELSD 2000ES (Alltech, China). The chromatographic column was Phenomenex Luna C18 (2) (4.6 × 250 mm, 5 µm) at 30 ºC. The mobile phase was composed of aqueous solution of 0.1 % formic acid (eluent A) and acetonitrile (eluent B), and the gradient program used was 10-15% B ( 0-5 min), 15-19 % B (5-25 min), 19-28 % B (25-32 min), 28-35 % B (32-57 min), 35-65% B (57-67 min), 65-10% B (67-69 min), and 10% B (69-75 min). The flow rate and the injection volume were 1.0 mL /min and 10 µL, respectively. The parameters of ELSD 2000 ES were set with the model of impactor off, drift tube temperature of 108 ºC and N2 gas pressure of 13.8 psi.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
