T Cell Activation and Proliferation

Check if the same lab product or an alternative is used in the 5 most similar protocols
T cells were purified from pooled lymph nodes and spleens of wild-type and LckS59A mice with Dynabeads Untouched Mouse T cell kits (Life Technologies) with a cell purity of over 90%. Purified cells were stimulated for 72 h with plate-bound anti-CD3 (145-2C11; American Type Culture Collection) with or without soluble anti-CD28 (37–51; American Type Culture Collection). Intracellular IL-2 and IFN-γ were measured as described (Mingueneau et al, 2008 (link)). Proliferation was measured using CFSE labelling. After 3 days, the absolute numbers of T cells having diluted CFSE were counted after adding Flow-Count™ Fluorospheres (Beckman Coulter).

Expansion and Stimulation of Purified CD4+ T Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
CD4+ T cells were purified from pooled lymph nodes and spleens with Dynabeads Untouched Mouse CD4+ T cell kits (Life Technologies) with a cell purity of over 95%. Purified CD4+ T cells were briefly expanded with plate-bound anti-CD3 (145-2C11; American Type Culture Collection) and soluble anti-CD28 (37-51; American Type Culture Collection). After 48 h of culture, CD4+ T cells were harvested and grown in the presence of IL-2 (5-10 U/ml) for 48 h prior to stimulation.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
