Serum sCXCL16 Quantification Protocol

Within 24 h of admission, 5 mL of whole blood was collected in dry tubes. Blood samples were then centrifuged at 2000 rpm for 5 min.
A multiplex kit “Human cytokine/chemokine magnetic bead Panel IV” for MAGPIX® multiplexing system for Luminex (EMD Millipore Corporation) was used to measure the sCXCL16 concentration in serum from the 76 patients in the matched groups. Each serum was assessed in duplicate at a 1/4 dilution according to the manufacturer’s instructions. The final concentration was used as the mean. The coefficients of variation [CVs] between each duplicate are presented in Supporting Information: Table I. The inter-assay CV (between the two plates) were calculated using standards concentrations (Supporting Information: Table II). CXCL16 standards were used to calculate a six-point five-parameter logistic standard curve and assess sCXCL16 levels with the Belyssa 1.0.19 software, and values below the lower limit of quantification (LLOQ) were replaced with an arbitrary value (LLOQ/2).
