Phenylhydrazine-Induced Anemia in Mice

129/SvImJ (wild-type), 129S4/SvJae (wild-type), or 129S4/SvJae-Pparαtm1Gonz/J female mice (The Jackson Laboratory) 6 – 8 weeks of age, randomized by weight, were pretreated with GW7647 (100 μg/kg) for 3 days (days −3 – −1). On day 0, the mice were injected with phenylhydrazine (PHZ) (60 mg/kg). GW7647 injection was continued during days 0 – 6. Whole blood samples were collected at each day of day 1 – 5, and day 7 and 9 for Complete Blood Count (CBC) analyses.
4 to 6 week old Nan/+ mutant mice (officially designated as Klf1Nan; http://www.informatics.jax.org/allele/MGI:1861107), obtained from the Jackson Laboratory, were injected with GW7647 (100 μg/kg) for 18 days. Mice were randomized by weight. Whole blood samples were collected every 3 days for CBC analyses.
All mouse procedures were approved by the Animal Care and Use Committees of the Massachusetts Institute of Technology (Cambridge, MA).

Murine Studies of VWF-FVIII Clearance

Check if the same lab product or an alternative is used in the 5 most similar protocols
VWF−/− and FVIII−/− mice on C57BL/6J and 129S4/SvJae backgrounds, respectively were obtained from the Jackson Laboratory (Sacremento, CA). These mice were crossbred to generate VWF−/−FVIII−/− double-knockout mice and bred for 10 generations to C57BL/6J background. MGL1−/− mice were also obtained from the Jackson Laboratory. Mice were fed ad libitum with standard chow (LabDiet, IPS United Kingdom). In keeping with previous murine studies of VWF-FVIII clearance, studies were performed in both male and female mice,19 (link)–24 (link) and data combined in order to improve power. Consequently, potential sex differences were not investigated in the study. All in vivo studies were performed in accordance with the Health Product Regulatory Authority, Ireland (AE19127/P060) and approved by the Animal Research Ethics Committee of The Royal College of Surgeons in Ireland and Trinity College Dublin (REC1585 and 260219, respectively). Where indicated, clearance studies were repeated in the presence or absence of (1) clodronate-induced macrophage depletion16 (link),17 (link) or (2) polyclonal goat anti-mouse MGL1/2 antibody (R&D systems, Minneapolis) as previously described.16 (link),17 (link),20 (link) Plasma FVIII:C levels were measured using a chromogenic assay (HyphenBiomed, France).

Geminin Conditional Knockout in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Geminin flox/flox (Gmnnfl/fl) mice (Shinnick et al., 2010 (link)) were maintained with a mixed genomic background of 129S4/SvJae and C57BL/6J (016913; Jackson Laboratory, Bar Harbor, ME), and Gdf9-cre mice (Lan et al., 2004 (link)) were maintained with C57/BL6 genomic background. Mutant mice were homozygous for the geminin floxed allele and heterozygous for Gdf9-cre (Gdf9-cre Gmnnfl/fl), and control mice were homozygous for Geminin floxed allele and Gdf9-cre negative (Gmnnfl/fl). Mice were housed in 12-h alternating light/dark cycles, with free access to water and food.
All experiments were conducted with the approval of the Animal Research Committee of the Institute of Zoology, Chinese Academy of Sciences. Mice were killed under standard protocols, and all efforts were made to minimize suffering.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
