Briefly, proteins were separated by SDS-PAGE and transferred to a nitrocellulose membrane, followed by detection using specific antibodies. The antibodies were used included rabbit monoclonal anti-survivin, rabbit polyclonal anti-phospho-STAT3 (Tyr705), and anti-cleaved-PARP (Asp214) from Cell Signaling Technology (Beverly, MA); mouse monoclonal anti-STAT3, rabbit polyclonal anti-ERa, and anti-HER2 from Santa Cruz Biotechnology Inc. (Santa Cruz, CA); and mouse monoclonal anti-β-actin from Sigma (St. Louis, MO). Blots were developed using horseradish peroxidase-conjugated secondary antibody and an enhanced chemiluminescence detection system (Amersham Life Science, Piscataway, NJ).
