Formic acid and methanol were obtained from Sigma–Aldrich(St. Louis, MO, U.S.A.) and Merck (Darmstadt, Germany), respectively. Ethanol and acetic acid were purchased from J.T. Baker (Phillipsburg, NJ, U.S.A.). Distilled deionized water was produced by a PURELAB purifier (ELGA, Marlow, U.K.). Standards for illegal drugs including 5-MeO-AMT, amphetamine (AM), methamphetamine (MA), mephedrone, para-methoxymethamphetamine (PMMA), 3,4-methylenedioxymethamphetamine (MDMA), methylone, 2-(4-chloro-2,5-dimethoxyphenyl)ethan-1-amine (2C-C), ketamine, nitrazepam, flunitrazepam (FM2), and lysergic acid diethylamide (LSD) were purchased from Cerilliant Corporation (Round Rock, TX, U.S.A.). Stock solutions of illicit drugs were prepared in MeOH. Drinks and powders for whole fat milk (Uni-President Enterprises Corporation), grape juice (Uni-President Enterprises Corporation), fruit and vegetable juice (Chou-Chin Industrial Co.), sugar-free green tea bags (Vitalon Foods Company), 3-in-1 instant coffee packs (Nestlé Company), matcha tea packs (Starbucks), and gelatin powder (Tehmag Foods Corporation) were obtained from local supermarkets. Postage stamps were purchased from Chunghwa Post Co., Taiwan.

Quantification of Illicit Drugs in Cough Syrups

Carbinoxamine, chlorpheniramine, and chlorpheniramine-d6 were purchased from TRC (North York, Canada). Individual solutions of 6-AM, 7-aminonitrazepam, 7-aminonimetazepam, amphetamine, benzoylecgonine, C6G, codeine, codeine-d6, ketamine, methamphetamine, mephedrone, 4-methylephedrine, 3,4-methylenedioxymethamphetamine hydrochloride, M3G, M6G, morphine, morphine-d6, and norketamine (each with a concentration of 1 mg mL−1) were purchased from Cerilliant (Round Rock, TX, USA). Three brands of cold syrups (Consrine, ssBuron, and Pabron) manufactured in Taiwan were purchased from local pharmacies. Consrine contains codeine phosphate (0.48 mg mL−1), chlorpheniramine (0.25 mg mL−1), dl-methyl ephedrine HCl (1.6 mg mL−1), and glycyrrhizic acid (0.25 mg mL−1), ssBuron contains codeine phosphate (0.5 mg mL−1), guaifenesin (3.33 mg mL−1), chlorpheniramine maleate (0.14 mg mL−1), and caffeine (1.33 mg mL−1), Pabron contains codeine phosphate (0.48 mg mL−1), dl-methyl ephedrine HCl (2.5 mg mL−1), and carbinoxamine maleate (0.4 mg mL−1). Formic acid and methanol (high-performance LC grade) (≥ 98%) were purchased from J.T. Baker (Pittsburgh, PA, USA). Ultrapure water (18.2 mΩ·cm) obtained from a Milli-Q ultrapure water system (Millipore, Billerica, MA, USA) was used in this study.

Three standards of NPSs, 4′-chloro-alpha-pyrrolidinopropiophenone (4-Cl-α-PPP, 1 mg, purity, ≥98%), 4′-Fluoro-α-pyrrolidinopentiophenone (4-F-α-PVP, 1 mg, purity, ≥98%), and 1-pentyl-3-(2-bromophenylacetyl)indole (JWH-249, 1 mg, purity, ≥98%) were purchased from Cayman Chemicals (Ann Arbor, MI, USA). Sodium azide (500 mg, purity, ≥99%) was purchased from Sigma-Aldrich (St. Louis, MO, USA).

Quantitative Analysis of Ketamine and Metabolites

ketamine dosing solution was purchased from Putney, Inc. (Portland, MA) and either used as directed or aseptically diluted using sterile saline to a final concentration of 25 mg/mL. The concentration of the ketamine dosing formulations was verified using LC-MS as described below. Atropine was purchased from Sigma Aldrich (St. Louis, MO) and prepared at a concentration of 15 mg/mL in sterile saline. The pH of the vehicle was acidified with dilute hydrochloric acid to increase solubility before being neutralized to pH 7.2 with dilute sodium hydroxide. The 2-PAM used was purchased from Sigma Aldrich (St. Louis, MO) and prepared at 50 mg/mL in sterile saline and sterile filtered. Midazolam (5 mg/mL) was purchased from Hospira (Lake Forest, IL) and used without modification.
The analytical reference standards for ketamine, norketamine, ketamine-d4, and norketamine-d4 in methanol were obtained from Cerilliant (Round Rock, TX). Multiple lots of K3EDTA rat plasma and brains were obtained from Bioreclamation (Westbury, NY). Purified water was collected from an in-house Labconco (Kansas City, MO) Water Pro Plus system. Glacial acetic acid was obtained from JT Baker (Center Valley, PA). Ammonium hydroxide was obtained from Sigma Aldrich (St. Louis, MO). LC-MS-grade water and acetonitrile were obtained from EMD (Billerica, MA). Formic acid was obtained from Thermo Scientific (Waltham, MA).
