Emodin-Loaded Nanoparticles for Cellular Responses

Measuring Merozoite Oxidative Stress

mROS was measured using MitoSOX Red (MedChemExpress, Shanghai, China) according to the manufacturer’s instructions. Briefly, after incubation of merozoites with different concentrations of drugs, merozoites from all groups were incubated with 5 μM MitoSOX for 15 min at 37 °C in the dark. After washing 3 times with warm DMEM, we detected the ROS intensity using a fluorescence spectrophotometer (Tecan, Infinite M200 PRO, Männedorf, Switzerland).

Fluorescent Reagents for Cell Analysis

4 kDa FITC-dextran was obtained from Maokang Biotechnology (Shanghai, China). Roswell Park Memorial Institute (RPMI) 1640 medium and Dulbecco’s Modified Eagle Medium (DMEM) was procured from Gibco (USA). Antibodies for Piezo1, Occludin and ZO-1 were procured from Proteintech (China). The following chemicals were obtained from Servicebio (China): fluorescein (FITC) Tunel Cell Apoptosis Detection Kit, Phosphate Buffered Saline (PBS) and Hank’s Balanced Salt Solution (HBSS). Fluorescent-conjugated antibodies for flow cytometry were procured from BD Bioscience (USA). Secondary antibodies conjugated with AlexaFluor-488 or Alexa Fluor 594, Annexin V-FITC /PI Apoptosis Detection Kit, JC-1 Mitochondrial Membrane Potential Assay Kit, SYBR qPCR Master Mix, Rhod-2 AM and Fetal Bovine Serum (FBS) were procured from Yeason (China). Yoda1 and MitoSOx Red were procured from MedChemExpress (USA). MitoTEMPO was acquired from Topscience (China). DMEM no Ca2+ was obtained from Meiluncell (China).

Fluorescence-based Mitochondrial and ROS Assays

Fluorescence-based measurements of mitochondrial membrane potential and intracellular ROS generation were performed as previously described 50 (link). In brief, following treatment, hepatocytes were incubated with the potentiometric JC-1 sensor (Mitochondrial Membrane Potential Detection Kit, #30001, Biotium). For ROS measurements, fresh liver sections or primary hepatocytes were incubated with the cellular ROS indicator DHE (#PD-MY 003, MedChemExpress), cytoplasmic ROS indicator H2DCFDA (#HY-D0940, MedChemExpress), and mitochondrial ROS indicator MitoSOX RED (HY-D1055, MedChemExpress) based on methods provided by manufacturers 51 (link). Fluorescence images were acquired with same laser output and gain for control and treated samples on a Zeiss 880 confocal microscope using a 100X oil objective 52 (link).

Ferroptosis-related Molecular Mechanisms

Anti-GAPDH (sc-47724), anti-β-tubulin (sc-5274), anti-β-actin (sc-47778), anti-CBS (sc-133154), anti-Nrf2 (sc-365949), and anti-GPX4 (sc-166570) antibodies were from Santa Cruz Biotechnology (Santa Cruz, CA). Lipofectamine 3000 (L3000), Lipofectamine RNAi MAX (13778), the lysosome-specific dye LysoTraker Red (L7528), and Alexa Fluor 488 goat anti-rabbit IgG (R37116) were purchased from Thermo Fisher Scientific (Waltham, MA). All other chemicals were purchased from Sigma-Aldrich (St. Louis, MO).
