Elemental Analysis of Potato Tissues

The concentrations of the following cationic macronutrients: calcium (Ca), potassium (K), magnesium (Mg) and micronutrients: iron (Fe), copper (Cu), manganese (Mn), zinc (Zn) in potato leaf and root tissues were analyzed through Inductively Coupled Plasma Optical Emission Spectroscopy (ICP-OES) as previously described.21 The same potato tissues used for qPCR analyses were oven-dried and 0.5 g of dried tissue was ashed at 510 °C for 9 h, allowed to cool, and digested in 10 ml of 1 N HNO3 for 1 h. The filtered supernatant was brought to volume (25 ml) and the intensities of atomic emissions at 396.847 nm for Ca, 766.491 nm for K, 279.553 nm for Mg, 238.204 nm for Fe, 327.395 nm for Cu, 257.610 nm for Mn, and 213.857 nm for Zn was measured on an ICP-OES System (Varian Vista Pro CCD Simultaneous ICP-OES attached to Varian SPS 5 Sampler Preparation System, Agilent, USA). Samples were diluted 1:100 in 1 N HNO3 prior to Ca, K, and Mg analyses. All containers used for ICP Spectroscopy analysis were acid-washed by soaking overnight in 1 N HNO3 before use.

Maize Yield and Grain Quality Analysis

Maize was harvested at the fully ripe stage and plants from border rows were harvested separately and were not used for further measurements. Grain yield was calculated at 14% of moisture content. Number of grains per ear and 1000 grain weight were determined, and samples for determination of grain protein and Zn concentration were taken. Analytical methods. Zn concentration was measured in seeds and harvested grains, and protein concentration was measured in grains. To obtain grain Zn concentration samples were air dried and milled, and 0.5 g of dry samples were microwave digested with 10 ml of HNO3 + 2 mL H2O2 (Milestone ETHOS UP, Milstone Inc., Shelton, USA). Grain Zn concentration was determined by inductively coupled plasma optical emission spectroscopy (Varian Vista-PRO CCD Simultaneous ICP-OES, Varian, Inc.). Protein concentration in grain was determined by nearinfrared spectroscopy (NIR Analizer, INSTALAB 600, Dickey John, USA).
