Cytokine Secretion Assay for T Cells

For measurement of cytokine secretion, human T cells (106 cells/mL) were seeded in anti-CD3/CD28 antibody-coated 24-well plates (Corning Inc., Amsterdam, The Netherlands) and treated with increasing concentrations of Pitavastatin or vehicle control. The optical density of each well was determined at 450 and 570 nm (Tecan Safire Microplate Reader, Tecan Group, Männedorf, Switzerland). The concentration of each cytokine was calculated by using a standard curve and the measured absorbance (n = 4 independent experiments).
