Ultra low attachment 6 well culture plates

Manufactured by Thermo Fisher Scientific

The Ultra Low Attachment 6-well culture plates are designed for culturing cells in a controlled, non-adhesive environment. These plates feature a specialized surface treatment that prevents cell attachment, promoting the formation of 3D cellular structures such as spheroids or organoids. The 6-well format provides a convenient platform for various cell culture applications.

Differentiating ENC Cells into Neurons

Check if the same lab product or an alternative is used in the 5 most similar protocols
ENC cells from the 11 day induction protocol were aggregated into 3D spheroids (5 million cells/well) in Ultra Low Attachment 6-well culture plates (Fisher Scientific, 3471) and cultured in Neurobasal (NB) medium supplemented with L-Glutamine (Gibco, 25030-164), N2 (Stem Cell Technologies, 07156) and B27 (Life Technologies, 17504044) containing CHIR99021 (3uM, Tocris Bioscience, 4423) and FGF2 (10nM, R&D Systems, 233-FB-001MG/CF). After 4 days of suspension culture, the spheroids are plated on Poly Ornithine/Laminin/Fibronectin (PO/LM/FN) coated dishes (prepared as described previously26 ) in Neurobasal (NB) medium supplemented with L-Glutamine (Gibco, 25030-164), N2 (Stem Cell Technologies, 07156) and B27 (Life Technologies, 17504044) containing GDNF (25 ng/ml, Peprotech, 450-10) and Ascorbic acid (100 uM, Sigma, A8960-5G). The ENC precursors migrate out of the plated spheroids and differentiate into neurons in 1-2 weeks. The cells were fixed for immunostaining or harvested for gene expression analysis at Day 25, Day 40 and Day 60 of differentiation.

Differentiation of Neural Crest Cells into Schwann Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
At day 11, NC cells were aggregated into 3D spheroids (5 million cells/well) in Ultra Low Attachment 6-well culture plates (Fisher Scientific, 3471) and cultured in Neurobasal (NB) medium supplemented with L-Glutamine (Gibco, 25030–164), N2 (Stem Cell Technologies, 07156) and B27 (Life Technologies, 17504044) containing CHIR (3 μM, Tocris Bioscience, 4423) and FGF2 (10 ng/ml, R&D Systems, 233-FB-001MG/CF) and NRG1 (10 ng/ml, R&D 378-SM-025). After 14 days of suspension culture, the spheroids were plated on Poly Ornithine/Laminin/ Fibronectin (PO/LM/FN) coated dishes (prepared as described previously (17)) in Neurobasal (NB) medium supplemented with L-Glutamine (Gibco, 25030–164), N2 (Stem Cell Technologies, 07156) and B27 (Life Technologies, 17504044) containing NRG1 (20 ng/ml, R&D 378-SM-025), FGF2 (10 ng/ml, R&D Systems, 233-FB-001MG/CF) and cAMP (100 mM, Sigma, D0260). The SC precursors migrate out of the plated spheroids and differentiate into SCs within 10 days. For long-term expansion, cells were cultured in Schwann cell medium (Sciencell, 1701 without FBS) on PO/LM/FN coated dishes. Cells were fixed for immunostaining or harvested for gene expression analysis at Day 25, Day 35, Day 50 and Day 100 of differentiation.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
