Gelatin-based Tissue Engineering Scaffold

Gelatin supplied from Nitta-Gelatin Ltd. (Japan headquarter) is a high-grade quality, low-endotoxin unit essential for diminishing immune rejection post-implantation. It is currently manufactured at Nitta-Gelatin Ltd. (India branch) and is certified halal, originating from a buffalo’s raw bone. Quercetin (QC) was purchased from Sigma-Aldrich (St. Louis, MO, USA) and utilized without further purification. Elastin was procured from the Faculty of Science and Technology, Universiti Kebangsaan Malaysia (FST, UKM) and Genipin was purchased from FUJIFILM Wako, Osaka, Japan. Pharmaceutical-grade solvents and reagents were used in this study and were used as received.
The location for this study was the Centre for Tissue Engineering and Regenerative Medicine (CTERM), Faculty of Medicine, and several tests were run at FKAB, UKM, and iCRIM UKM. The study design was approved by the Universiti Kebangsaan Malaysia Research Ethics Committee (UKM PPI/111/8/JEP-2021-301).

Quercetin and Pluronic F127 Formulation

Quercetin (QC) and Pluronic F127 (Poloxamer 407; PF-127) were purchased from Sigma-Aldrich (Karnataka, India). Olive oil and Tween 80 were purchased from Loba Chemie (Mumbai, India). Phospholipid (Phospholipon® 90 G; PC) was kindly gifted from Lipoid (Steinhausen, Switzerland). Acetone (purity > 99%), ethanol (95%) and n-hexane (purity ≥ 97%) were purchased from Sigma-Aldrich (Steinheim, Germany). All other chemicals and solvents were in analytical grades.

Cytotoxicity Evaluation of Graphite-Based Nanomaterials

The human dermal fibroblast cells (HDF) as a normal cell, isolated from human newborn foreskins at 1 month of age that underwent routine circumcision in Amirkola Children’s Hospital (Babol, Iran).

Breast Cancer Cell Line Culture Protocol

MCF-7 human breast cancer cell line was obtained from American Type Culture Collection (ATCC) (Manassas, VA). Dulbecco's Modified Eagle's Medium (DMEM) High Glucose was purchased from Hy Clone Thermo Scientific. Fetal bovine serum (FBS) was obtained from Gibco-Life Technologies. Penicillin/streptomycin and 0.25% (w/v) trypsin-0.1% (w/v) Ethylenediaminetetraacetic acid (EDTA) were purchased from Solarbio (Beijing Solarbio Science and Technology, China). 3-(4, 5-dimethyl-thiazol-2-yl)-2, 5-diphenyl-tetrazolium bromide (MTT) and dimethylsulfoxide (DMSO) were purchased from Sigma (USA). Culture flasks and dishes were from Corning (Corning, NY, USA). Phosphate buffer saline (PBS) was obtained from the Iranian Research Organization for Science and Technology (IROST). Polyvinylpyrrolidone (PVP K90, 1,200,000 Daltons), agarose (AG) pharmaceutical grade type I low EEO (suitable for hydrogel preparation), hydroxyapatite (HAp) (Ca 10 (PO 4 ) 6 (OH) 2, 1004.6 g/mol), and quercetin (QC) (3,3′,4′,5,7pentahydroxyflavone) were purchased from Sigma-Aldrich. Span 80 and paraffin oil were obtained from Merck and Carlo Erba Reagents, respectively. Glyoxal and Polyvinyl alcohol (PVA) were obtained from Sigma-Aldrich.

Topical Antimicrobial and Anti-Inflammatory Formulation

Quercetin (QC) and polyvinyl alcohol (PVA) Mwt 125,000 were both purchased from Sigma Aldrich, Germany. Essential oils (tea tree oil, neem oil) were both purchased from Alpha Pharmaceutical Co., Cairo, Egypt. All analytical grade solvents were purchased from Fisher Scientific UK. Disodium hydrogen phosphate, and potassium dihydrogen phosphate were purchased from El Nasr chemical Co., Cairo, Egypt. Propionibacterium acne (P. acne) ATCC 6919, horse blood agar, 3T3 CCL92 skin fibroblastic cells, trypsin, antibiotics (penicillin, streptomycin), barium chloride dehydrate, sulfuric acid, hydrogen peroxide, and neutral red, were all purchased from Sigma Aldrich Co., UK as previously mentioned in the work conducted by (Amer et al., 2020). Dulbecco modified eagle's medium (DMEM), and FBS (Fetal Bovine serum 10%) were both purchased from Gibco by life technologies, Thermo Fisher Scientific, USA.
Mueller Hinton agar, brain heart infusion, and the anaerobic indicator were purchased from Thermo Scientific, Oxoid, Anaerogen, UK. Industrial methylated spirit 70% (IMS) was purchased from Atom Scientific, UK. Panthenol ® cream was purchased from a local pharmacy (Sherryland Pharmacy) where it was manufactured by the Nile Co. for Pharmaceuticals and Chemical Industries, Egypt.
