Gastric Cancer Tissue Immunohistochemistry

Paraffin-embedded sections of 5 paired gastric cancer and adjacent normal tissues were collected from Heilongjiang Provincial Hospital. Each patient signed a written informed statement. This study gained the approval of the Ethics Committee of Heilongjiang Provincial Hospital (2020061). All specimens were fixed through 10% formalin for 48 h and sectioned into 5 μm thickness. The sections were incubated by primary antibodies against APOD (1/100; ab108191; Abcam, USA), CTLA4 (1/100; ab237712), CXCR4 (1/100; ab197203), DKK1 (1/100; ab109416), INHBA (1/100; ab97705), NPR1 (1/100; ab40817), PENK (1/100; ab22619), PROC (1/100; ab17771), RBP4 (1/100; ab188230), S100A12 (1/100; ab196740), and STC1 (1/100; ab229477) overnight at 4°C and incubated by HRP-labeled secondary antibodies for 30 min at room temperature. Afterwards, the sections were stained by hematoxylin and investigated under a microscope.

Antibody Generation and Characterization

A rabbit polyclonal antibody against the human ABCC11 protein (anti-ABCC11 antibody (09YT)) was raised by immunizing rabbit with synthetic oligo-peptides as described above. Other anti-ABCC11 antibodies—such as H-215 (sc-20969) and M8I-74 (ab91452)—were purchased from Santa Cruz Biotechnology, Inc. (Santa Cruz, CA, USA) and Abcam Inc. (Cambridge, MA, USA), respectively. Antibodies against α-tubulin (ab15246) and apolipoprotein D (ab108191) were obtained from Abcam Inc. Antibody against enhanced green fluorescent protein (EGFP) (A11122) was purchased from Life Technologies (Tokyo, Japan). A donkey anti-rabbit immunoglobulin G (IgG)-horseradish peroxidase (HRP)-conjugated antibody (NA934V) and a goat anti-rat IgG-HRP-conjugated antibody (NA935V) were purchased from GE Healthcare UK Ltd. (Buckinghamshire, UK). All the other chemicals used were commercially available and of analytical grade.

Immunohistochemical Analysis of Gastric Tumor and Liver Metastasis

The study was reviewed and approved by the Ethics Committee of The Second Affiliated Hospital of Wannan Medical College (Wuhu, China) and the ethical number is WYEFYLS2023138. The study was conducted in compliance with the principles of the Declaration of Helsinki (as revised in 2013). All participants provided full written informed consent.
The case inclusion criteria included age 18 years or above; newly diagnosed primary gastric tumor and Li confirmed by histology; without other treatments. The exclusion criteria included history of other malignancies. Three patients with newly diagnosed primary gastric tumor and Li who attended The Second Affiliated Hospital of Wannan Medical College were invited to participate in this study. All patients provided full written informed consent. We obtained three pairs of fresh tissue samples which contained primary gastric tumor tissues and Li tissues from these patients.
IHC was performed as previously described (26 (link)) on clinical specimens of gastric orthotopic tumor and Li using the primary antibodies against APOD (1:100, ab108191, Abcam, Cambridge, UK), JUN (1:100, ab40766, Abcam), CXCL5 (1:100, ARG66029, Arigo, Hsinchu, Taiwan, China), TNFAIP3 (1:100, ab270264, Abcam), IL7R (1:100, AG2272, Beyotime, Shanghai, China) and CD94 (1:100, ab235441, Abcam).
