Hepatocyte Cell Line Cultivation Protocol

Undifferentiated human HepaRG cells (HPR101) were purchased from Biopredic International (Rennes, France) and human normal immortalized THLE2 and hepatoma Huh7 cells were obtained from American Type Culture Collection. William’s E medium with no glutamine, bronchial epithelium growth medium (BEGM) medium with complete supplement, Dulbecco’s modified Eagle medium (DMEM), fetal bovine serum (FBS), fibronectin, phosphate buffered saline (PBS), L-glutamine, penicillin/streptomycin, and trypsin-EDTA were purchased from Thermo Fisher Scientific (Rockford, IL, USA). Collagen type I was purchased from Advanced Biomatrix (San Diego, CA, USA). Insulin, hydrocortisone hemisuccinate, dimethyl sulfoxide (DMSO), methanol, acetonitrile anhydrous, CHIR, 30% wt H2O2 solution, and agarose (low gelling temperature) were purchased from Sigma-Aldrich (St. Louis, MO, USA). Chlorzoxazone, tamoxifen, isoniazid, bromobenzene, and APAP were acquired from Sigma-Aldrich (USA).

HPLC Analysis of Diverse Organic Solutes

The test solutes used in this study were obtained from various suppliers. Toluene, propylbenzene, butylbenzene, biphenyl, phenol, benzoic acid, aniline, N,N-dimethylaniline, caffeine, o-cresol, p-cresol, m-cresol, phloroglucinol, bromobenzene, chlorobenzene, nitrobenzene, anisole, naphthoic acid, acetophenone, 2,4-dimethylphenol, 2,6-dimethylphenol, p-nitrophenol, o-nitrophenol, m-nitrophenol, uracil, naproxen, ibuprofen, nifedipine, and bupropion were acquired from Sigma–Aldrich. Benzaldehyde, naphthalene, and benzyl alcohol were purchased from Thermo Fisher Scientific. All chemicals were used as received without any further purification. Individual samples were prepared in methanol (Thermo Fisher Scientific) as the solvent at a concentration of 10−2 mM; the mixture containing the various components was prepared in methanol at a concentration of 2 mM each. Samples were filtered through a 0.45 μm membrane filter before injection into the chromatograph. HPLC-grade methanol from Thermo Fisher Scientific was used as the mobile phase modifier. Food-grade CO2 was purchased from PRAXAIR and used as the mobile phase.

Hydrogenolytic Deoxyhalogenation of Haloaromatic Compounds

Raney Ni (RTH-3110) and 5% Pd/C catalyst used in this study were purchased from Dalian Tongyong Chemical Co., Ltd., Liaoning, China. The weight percentage of nickel and aluminum in the catalyst were more than 90% and less than 7%, respectively. Raney Ni and Pd/C catalyst are not pre-treated before all experiments, and they were kept in water-sealing storage and hermetical desicator, respectively.
All HACs including fluorobenzene (FB), chlorobenzene (CB), bromobenzene (BB), iodobenzene (IB), 4-fluorophenol (4-FP), 4-chlorophenol (4-CP), 4-bromophenol (4-BP), and 4-iodophenol (4-IP) used in experiments were purchased from Sigma-Aldrich with a minimum purity of 98%. The other reagents such as EtOH, NaOH, NaF, NaCl, NaBr, and NaI are analytical grade and supplied by Sinopharm Chemical Reagent Co., Ltd., Shanghai, China. Deionized water was used in the reaction. The purity of hydrogen and nitrogen used in the experiments is more than 99.99%.

Hepatoprotective Efficacy of Silymarin

The antibodies against Caspase-3, Cyclooxygenase-2 (COX-2) and Necrosis factor kappa B (NF-κB) for Western blotting were purchased from Taurus Inc., India. Commercial diagnostic kits for aspartate amino transferase (AST), alanine amino transferase (ALT), alkaline phosphatase (ALP), total and direct bilirubin, total cholesterol, high density lipoprotein (HDL), total protein and albumin were procured from AutoSpan Diagnostics Ltd., Surat, Gujarat, India. All the other chemicals and reagents used were acquired locally and of analytical grade.
