NK Cell Stimulation Assay

104 purified splenic Ly49Hlo or
Ly49Hhi NK cells were stimulated for 4 hours in RPMI
containing 10% fetal bovine serum with 20 ng/mL recombinant mouse IL-12
(R&D Systems) plus 10 ng/mL IL-18 (MBL), 10 ng/mL PMA (Sigma) plus 1
g/mL Ionomycin (Sigma), or 25 g/mL of plate-bound anti-mouse NK1.1 (PK136,
BioLegend) or anti-mouse Ly49H (3D10, BioLegend). Cells were cultured in
media alone as a negative control.
