Polysulfide donor Na₂S₄ was purchased from Dojindo Molecular Technologies Dojindo (Kumamoto, Japan). Cisplatin and NaHS were bought from Sigma-Aldrich (St. Louis, MO, USA). GYY4137 was synthesized as we previously depicted [72 (link)]. A selective and reversible inhibitor of IKKβ (IKK-2), SC-514, was procured from Santa Cruz Biotechnology Inc. (Santa Cruz, CA, USA). A selective STAT3 inhibitor HO-3867 was obtained from Medchem express (Princeton, NJ, USA). Antibodies against IL-6, COX-2, F4/80, Biotin, Lamin B1, and β-actin, and the secondary antibodies, were purchased from Santa Cruz Biotechnology Inc. (Santa Cruz, CA, USA). Antibodies against TNF-α, IL-1β, phosphorylated and total IκBα, NF-κB, phosphorylated and total STAT3, and phosphorylated and total IKKβ were obtained from Abcam (Cambridge, MA, USA). Dulbecco’s modified Eagle’s medium (DMEM), streptomycin/penicillin, trypsin, and fetal bovine serum (FBS) were provided by Hyclone Laboratories (South Logan, UT, USA). The radio-immunoprecipitation assay (RIPA) lysis buffer was purchased from Thermo Fisher Scientific Inc. (Waltham, MA, USA). Goat anti-mouse IgG (H+L) Highly Cross-Adsorbed Alexa Fluor 594 Secondary Antibody was procured from Invitrogen (Carlsbad, CA, USA). The specific primers used in this study were synthesized by Integrated DNA Technologies (Singapore).

Tris, glycine, NaCl, SDS, mannitol, bovine serum albumin (BSA), sodium hydrosulfide (NaHS), β-glycerophosphate (β-GP), ascorbate, and propargylglycine (PAG) were purchased from Sigma-Aldrich (St Louis, MO, USA). HO-3867 and LY3000328 (LY) were from Medchem express, LLC (Princeton, NJ, USA). Dulbecco’s modified Eagle’s medium (DMEM), fetal bovineserum (FBS), streptomycin/penicillin and trypsin were obtained from Hyclone Laboratories (South Logan, UT, USA). The RIPA buffer was purchased from Thermo Fisher Scientific Inc (Waltham, MA, USA). The Bradford colorimetric protein assay kit (Rockford, IL, USA) was used for protein quantification. The calcium, phosphorus, and alkaline phosphatase (ALP) kits were purchased from Jiancheng Bioengineering Co (Nanjing, China).

The human colorectal cancer cell lines HCT116 and SW480 (Obtained from Zhong Qiao Xin Zhou Biotechnology Co., Ltd., China) were cultured in DMEM (Gibco BRL, Grand Island, NY) complete medium containing 10%, FBS and 1% penicillin/streptomycin (Zhong Qiao Xin Zhou Biotechnology Co., Ltd., China) and incubated in an incubator containing 37 °C and 5% CO2. Fresh medium was exchanged every 2 days, and when the cell density reached 80–90%, the cells were digested with trypsin solution for subculture or plate laying. HO-3867 (10 µmol/l, MedChemExpress LLC, China) is a STAT3 inhibitor used to inhibit the expression of STAT3 [32 (link)].
