Gpx activity assay kit

Glutathione peroxidase (GP_X) activity, thioredoxin reductase (TR) activity, total superoxide dismutase (SOD) activity, total antioxidant capacity (T-AOC), malondialdehyde (MDA) content, and total protein content of the muscle samples were determined in accordance with the instructions of a GPx activity assay kit (colorimetric method), TR activity assay kit (colorimetric method), SOD activity assay kit (WST-1 cell proliferation reagent method), T-AOC assay kit (2,2′-azino-bis 3-ethylbenzothiazoline-6-sulfonic acid method), MDA assay kit (thiobarbituric acid method), and total protein quantitative assay kit (Bradford method) (Nanjing Jiancheng Bioengineering Institute, Nanjing, China), respectively. The reactive oxygen species (ROS) content of the muscle samples was determined in accordance with the instructions of an ROS assay kit (enzyme-linked immunosorbent assay methods) (Shanghai Jianglai Biological Technology Co., Ltd., Shanghai, China).

Fresh L4–L6 anterior horn tissue from rats (n = 5) was accurately weighed. Normal saline was added to the spinal cord tissue at a ratio of 9:1. After homogenization and centrifugation, the protein concentration in supernatant was measured. The iron level, malondialdehyde (MDA) content, glutathione (GSH) amount, and glutathione peroxidase (GPX) activity were determined using an iron assay kit (Jiancheng, Nanjing, China, A039-2-1), an MDA assay kit (Jiancheng, A003-1-2), GSH assay kit (Jiancheng, A006-2-1), and a GPX activity assay kit (Jiancheng, A005-1-2).

GPX activity in spinal cord tissue was determined using a GPX activity assay kit (A005, Nanjing Jiancheng Bioengineering Institute) as instructed by the manufacturer. OD values for each group were measured at 412 nm, and GPX activities were calculated.
