Transmission Electron Microscopy of MEL-XY3 Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
MEL-XY3 cells, MEL-XY3 SUR cells, and MEL-XY3 SUR cells at 2 and 7 days after PLX4032 removal were analyzed by TEM. Cells were harvested, fixed for 4 h at 4°C in 2.5% glutaraldehyde, post-fixed in 1% (v/v) osmium tetroxide. Subsequently, samples were embedded in Durcupan Epoxy resin (Sigma-Aldrich; Merck KGaA), and ultrathin sections (70-90 nm) were mounted on copper grids and stained with uranyl acetate. Stained samples on grids were visualized using a Zeiss EM 109T TEM (Carl Zeiss AG), and digital micrographs were captured with a Gatan ES1000W digital camera (Gatan, Inc.) in the LANAIS-MIE facility (Buenos Aires, Argentina).

Transmission Electron Microscopy of EVs

Check if the same lab product or an alternative is used in the 5 most similar protocols
To verify the presence of intact EV, the preparations were analyzed using transmission electron microscopy (TEM). The EV suspension was fixed in nickel grids for TEM with carbon (200 mesh) (Agar Scientific Ltd., Stansted, Essex, UK), with 2% glutaraldehyde, 4% formaldehyde, and 5% sucrose in PBS; washed three times with ultrapure water; stained with 3% uranyl acetate; allowed to dry for at least 30 min; and examined under a transmission electron microscope (Zeiss EM 109T equipped with Gatan ES1000W digital camera).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
