Extraction and Characterization of Garlic Trypsin

Chemicals; trypsin (bovine pancreatic trypsin), Nα-benzoyl-DL-arginine-p-nitroanilide (BAPNA), phenylmethylsulfonyl fluoride (PMSF), Polyvinylpyrrolidone (PVP), acrylamide, bis-acrylamide, Tetramethylethylenediamine (TEMED), ammonium persulfate and Sodium Dodecyl Sulfate (SDS), acrylamide, bis-acrylamide, TEMED, ammonium persulfate and SDS were obtained from Sigma-Aldrich. All other reagents and chemicals used were of analytical grade.

PA Hydrogel Preparation and Functionalization

PA hydrogel with a nominal elastic modulus of 1050 Pa was prepared as previously described [26] (link)[27] (link)[28] (link)[29] (link)[30] (link). Briefly, 0.075 ml of 40% acrylamide (Sigma-Aldrich), 0.05 ml of bis-acrylamide (Sigma-Aldrich), 0.1 ml of 10 × PBS (Solarbio, Beijing, China), 0.05 ml of 1% tetramethylethylenediamine (Sigma-Aldrich), 0.05 ml of 1% potassium persulfate (Sigma-Aldrich), and 0.675 ml of ultra-pure water were mixed to form a solution with a final concentration of 3% acrylamide and 0.1% bis-acrylamide. 100 μl mixture was added onto the hydrophilic-treated glass-bottom dish, covered with an 18 mm diameter circular coverslip to allow the hydrogel to be polymerized for 10 min, and rinsed with ultra-pure water overnight. The PA hydrogel surfaces were then conjugated to N-succinimidyl acrylamidohexanoic acid (VWR, Radnor, PA, USA) and activated for 15 min by 306 nm ultraviolet exposure and Irgacure 2959 (Sigma-Aldrich) catalyst immediately before coating with 100 μg/ml rat tail collagen I (Shengyou Biotechnology, Hangzhou, China) overnight.

Characterization of Prx Oxidation Profiles

Scientific (San Diago, USA), trifluoroacetic acid (TFA), formic acid (FA) and HPLC grade acetonitrile from Merck (Darmstadt, Germany) and HPLC grade water from J. T. Baker (PA, USA). Tris-HCl was from Duchefa (Haarlem, the Netherlands) and NP-40 from Amnesco (Ohio, USA).

Extraction and Antimicrobial Evaluation of Jatropha curcas Seed Cake

Jatropha curcas seed cake was obtained from Instituto Fazenda Tamanduá (Paraíba, Brazil), grounded in a coffee grinder and passed through a 1-mm-mesh screen. Azocasein, bovine pancreatic trypsin, bovine pancreatic chymotrypsin, bovine serum albumin (BSA), soybean trypsin inhibitor (SBTI), Nα-benzoyl-D,L-arginina-p-naftilamida (BANA), 4-(dimethylamino)cinnamaldehyde (DMACA), ethylenediaminetetraacetic acid (EDTA), phenylmethylsulfonyl fluoride (PMSF), L-cysteine and Nα-benzoyl-D,L-arginine 4-nitroanilide hydrochloride (BAPNA), sodium dodecyl sulfate (SDS), molar mass markers, acrylamide, bis-acrylamide, dithiothreitol (DTT) were purchased from Sigma-Aldrich (St. Louis, MO, USA). Chromatographic matrixes were from GE Healthcare. All other chemicals and reagents used were of analytical grade.

two major protein compounds. Two strips corresponding to the marker scale in
the range of 140 to 260 and 50 kDa were cut out for identification.

Photofunctionalisable Hydrogel Fabrication

Microscope glass slides were bought from VWR. These materials and chemicals were used to fabricate photofunctionalisable hydrogel films on glass substrates.
Materials used to make fluidic flow cells consisted of 3 mm thick transparent poly(methyl methacrylate) (PMMA) sheets, bootlace ferrules (211-4252), and nitrile O-rings (01-08-01801), and were purchased from Fred Aldous, RS Components, and Ashton Seals, respectively.
