Antioxidant and Neuroprotective Effects of HA and Seleninic Acid

Hydrogen peroxide solution (30 wt% in water) was obtained from Sinopharm Chemical Reagent Co., Ltd. 1,1-Diphenyl-2-picrylhydrazyl (DPPH) was purchased from Shanghai Macklin Biochemical Co., Ltd. Anti-GFAP, anti-NF200, and anti-NeuN primary antibodies were obtained from Abcam (Cambridge, UK). Anti-CD44 and anti-caspase-3 antibodies were purchased from Cell Signaling Technology (Danvers, MA, USA). The dialysis membrane was obtained from Greenbird Technology Co., Ltd (Shanghai, China). The remaining compounds were obtained from commercial providers and used without any further manipulation.

Histological Analysis of Intestinal Mucin

After the ileal and colonic tissues were fixed in 4% paraformaldehyde (Servicebio Technology, Wuhan, China) and dehydrated, the paraffin-embedded sections were dewaxed, stained with hematoxylin and eosin (H&E) (Servicebio Technology, Wuhan, China), and visualized using a microscope (Nikon, Tokyo, Japan). For the immunohistochemical evaluation of mucin MUC2 expression, the colonic sections were subjected to antigen repair, and 3% hydrogen peroxide solution (Sinopharm, Beijing, China) was added to block endogenous peroxidase activity. The tissues were sealed with 3% BSA (Servicebio Technology, Wuhan, China) at room temperature for 30 min, and then incubated with a primary antibody overnight at 4 °C. After incubation with a secondary antibody, the sections were stained with DAB (Servicebio Technology, Wuhan, China) and hematoxylin, and observed under a microscope.

Analytical Techniques for Biochemical Characterization

The formic acid, acetonitrile, and methanol used for high-performance liquid chromatography (HPLC) analysis were chromatographic grade and purchased from Merck KGaA (Shanghai, China). The ethanol, anthrone, sulphuric acid, glucose, DNS (3,5dinitrosalicylic acid), phenol, hydrochloric acid, sodium carbonate, sodium dihydrogen phosphate, disodium hydrogen phosphate, catechol, guaiacol, and hydrogen peroxide solution were all analytical purity (AR) and obtained from Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China). The bovine serum albumin (BSA), Coomassie Bright Blue G-250, Foline-phenol, and gallic acid were purchased from Yuan-ye Bio-Technology Co., Ltd. (Shanghai, China).
The ultraviolet spectrophotometer (UV-1800) was purchased from Shimadzu Co., Ltd. (Kyoto, Japan). The analytical balance (XS 250) was obtained from METTLER TOLEDO (Columbus, OH, USA). The high-speed centrifuge (2-16R) was purchased from HENGNUO Instrument Equipment Co., Ltd. (Changsha, Hunan, China).

Immunohistochemical Analysis of Ovarian IL-11 and IL-11Rα

The ovary tissues were washed with PBS and soaked in 4% paraformaldehyde for more than 24 h, dehydrated with ethanol, and embedded into paraffin blocks. The paraffin blocks were cooled in the freezer, sliced into 4 μm thick slices, and mounted on adhesion-treated slides. After deparaffinization, xylene was removed with a graded series of alcohols. Next, the slides were placed into a citric acid buffer (PH 6.0) for antigen retrieval in a microwave and endogenous peroxidase was blocked by a 3% hydrogen peroxide solution (Sinopharm, Beijing, China, Cat#10011208). After blocking with 3% BSA to decrease background staining, the treatment sections were incubated with primary antibodies against IL-11 and IL-11Rα (Table S1) and the negative controls were incubated with 3% BSA overnight at 4 °C. After three washes in PBS, the sections were incubated with the appropriate secondary antibodies (Table S1) for 50 min at room temperature and then stained with 3,3′-diaminobenzidine (DAB) solution (Servicebio, Wuhan, China, Cat#G1212). Subsequently, the sections were restained with hematoxylin (Servicebio, Wuhan, China, Cat#G1004) for 3 min and then dehydrated and sealed. Images were observed and captured under a microscope (Nikon, Tokyo, Japan).

Histopathological Evaluation of Acute Lung Injury

The fixed lung samples were embedded in paraffin as previously described [26] (link), and sectioned serially at 5 μm slices by using a rotary microtome (Leica, Nussloch, Germany). Subsequently, the samples were stained with hematoxylin and eosin (H&E) [27, (link)28] (link). Finally, the stained sections were observed and photographed under a BX53 microscope (Olympus, Tokyo, Japan). According to previous studies [29] (link), the scoring system was used to estimate the severity of acute lung injury as shown in Table 1.
Immunohistochemistry. For immunohistochemistry, the lung tissue sections were permeabilized in a 3% hydrogen peroxide solution (Sinopharm, Shanghai, China) for 15 min after antigen repair. Subsequently, the sections were blocked with 1% bovine serum albumin (BSA) (Sangon Biotechnology, Shanghai, China) for 15 min, and incubated with primary antibodies against HDAC6 (1:100, ABclonal Biotechnology, Wuhan, China) and NLRP3 (1:100, Affinity Biosciences, Changzhou, China) overnight at 4°. On the next day, the sections were incubated with the secondary antibody (1:500, ThermoFisher Scientific, Pittsburgh, PA, USA) at 37° in a humid chamber for 1 h. Afterward, images were observed and captured under a microscope (BX-53, Olympus).
