Caspase-Cleaved Cytokeratin 18 ELISA

Check if the same lab product or an alternative is used in the 5 most similar protocols
ELISA was performed to analyze the levels of caspase-cleaved cytokeratin 18 (cCK18) using the M30-Apoptosense ELISA kit (Peviva Ab, Bromma, Sweden). Cells (5000 cells/well) were seeded in 96-well plates and treated with 10 μM lenvatinib or DMSO for 24 h. Subsequently, the cells were lysed in polyoxyethylene octyl phenyl ether (Wako) and analyzed according to the manufacturer’s instructions. The experiments were repeated three times.

Immunocytochemistry of Liver Cells

The cultured cells were fixed and stained as previously described49 (link). Primary antibodies were as follows; mouse anti-human Hnf4a (PPMX), goat anti-human Foxa3 (Santa Cruz), goat anti-mouse albumin antibody (Bethyl Laboratories), mouse anti-mouse E-cadherin (Abcam), mouse anti-mouse cytchromeP450 1A2 (Abcam) at 1:200 respectively. The sections were incubated for two hours with secondary antibodies; Alexa 488-conjugated rabbit anti-goat IgG or goat anti-mouse IgG at 1:1000. After washing, the stained sections were covered with Vectashield mounting medium with DAPI (Vector Laboratories).
Spheroids were first embedded in iPGel (NIPPON Genetics) and fixed with 4% PFA overnight, then embedded in paraffin and cut in 5 μm thick. After deparaffinization and antigen retrieval using a Target Retrieval Solution (Dako), nonspecific binding was blocked with 1% bovine serum albumin (Sigma Aldrich) dissolved in 0.1% Polyoxyethylene Octylphenyl Ether (Wako) in PBS. The primary and secondary antibodies reactions were carried out in the same way for cultured cells.

Density-gradient and Gel Chromatography Purification of SWCNTs

Check if the same lab product or an alternative is used in the 5 most similar protocols
Metallic SWCNTs (Metal, bandgap: 0 eV) and semiconducting SWCNTs with a diameter of 1.4 nm (Semi. #1, bandgap: 0.7 eV) were prepared by the density-gradient sorting method from SWCNTs produced by the arc discharge method (Arc SO, Meijyo Nano Carbon Co.). 21 Semiconducting SWCNTs with an average diameter of 1.0 nm (Semi. #2, bandgap: 1.0 eV) and (6,5) SWCNTs (bandgap: 1.3 eV) were purified by the gel chromatography 22, 27 from raw soot of eDIPS (EC1.0, Meijyo Nano Carbon Co.) and CoMoCAT (SG65, Sigma-Aldrich), respectively. Each sample was filtered through a PTFE membrane (Omnipore 0.2 m PTFE Membrane, Merck Millipore Ltd.), and all surfactants on SWCNTs were replaced by Triton (polyoxyethylene octylphenyl ether, Wako Pure Chemical Industries, Ltd.). Then, the SWCNTs in 1% Triton solution were filtered through a polycarbonate membrane filter (Whatman Nuclepore Track-Etched membrane 25 mm 0.2 m) to form a thin film.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
