Quantifying IL-6 and IL-8 in Cell Cultures

Check if the same lab product or an alternative is used in the 5 most similar protocols
Concentrations of IL-6 and IL-8 in cell-culture supernatants were measured using IL-6 and IL-8 DuoSet ELISA kits (R&D Systems Europe, Abingdon, UK) according to the manufacturer’s instructions.33 (link) The limits of detection for the IL-6 and IL-8 ELISA kits were 9.38 and 31.3 pg/mL, respectively. Levels of IL-6 and IL-6-inducible CXCL1 (KC) in BALF were measured using commercial mouse IL-6 and KC ELISA kits (R&D Systems Europe, Abingdon, UK). The limits of detection for the mouse IL-6 and KC ELISA kits were 1.8 and 2.0 pg/mL, respectively.

Quantification of IL-8 Cytokine Levels

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cytokine concentrations were measured from aliquots of cell-free culture supernatants using commercially available IL-8 ELISA kits from Sanquin (Amsterdam, The Netherlands) for the cell lines (assay range = 1-240 pg/ml), and IL-8 duoset ELISA kits from R& D Systems Europe (Abingdon, UK) for NECs (assay range = 31.3-2000 pg/ml).

Measuring IL-6 and IL-8 in Chondrocytes

Check if the same lab product or an alternative is used in the 5 most similar protocols
The levels of IL-6 and IL-8 in culture supernatants from primary and immortalized articular chondrocytes after 24-hour treatment with IL-1β were determined with the human IL-6 and IL-8 DuoSet ELISA Kit (DY206 and DY208, respectively; Bio-Techne R&D Systems, UK), following the manufacturer's instructions. Absorbance measurements were performed using a NanoQuant Infinite M200 microplate reader (Tecan Ibérica Instrumentación, Spain). Data were expressed as released picograms per ml of culture supernatant. Working range was between 9.38 to 600 pg/ml for IL-6 and 31.25 to 2,000 pg/ml for IL-8.

Quantification of Secreted IL-8 by ELISA

Check if the same lab product or an alternative is used in the 5 most similar protocols
Secreted IL-8 protein levels were quantified with IL-8 DuoSet ELISA Kit (R&D Systems, Minneapolis, MS, USA) according to the manufacturer’s instructions. Supernatants were collected after 72 h of transfection, centrifuged and stored at − 20 °C until use. 3,3′,5,5′-Tetramethylbenzidine (TMB, Sigma-Aldrich) was used as visualizing reagent, substrate reaction was stopped with 1 M H2SO4. Optical density was measured with Epoch microplate spectrophotometer (BioTek, Winooski, VT, USA) at the wavelength of 450 nm.

Quantification of IL-6 and IL-8 Secretion

Check if the same lab product or an alternative is used in the 5 most similar protocols
HDMECs were transfected with si-control and si-NRP1 for 72 hours. Media was changed 24 hours after transfection and cells were cultured for an additional 48 hours. Cell culture supernatants were collected and analyzed with human IL-6 and IL-8 DuoSet ELISA kit following the manufacturer’s instructions (R&D System, UK; cat# DY208). Results were expressed as means of three independent experiments with triplicate samples.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
