In Vitro Metabolic Profiling of 7,8-Dihydroxyflavone

Nicotinamide adenine dinucleotide phosphate sodium salt (NADP+) and glucose-6-phosphate barium salt (G6P) were purchased from Reanal (Budapest, Hungary). Glacial acetic acid (99.5 v/v%) was obtained from Fluka (Bucharest, Romania). Stock solutions of DHF, site markers, substrates, and products (2000 μM each) were prepared in dimethyl sulfoxide (spectroscopic grade; Fluka) and stored at −20°C.

CD-1 male mouse liver microsomes pooled from 6424 mice, lot#2010017, and P450 contribution of 0.671 nmol/mg, were purchased from Sekisui Xenotech (Kansas City, KS, USA). Internal standard acenocoumarol was purchased from Toronto Research Chemicals (Toronto, ON, Canada). ACD/ChemSketch 2017.2.1 software (Toronto, ON, Canada) was used for rendering structures of molecules.

Quantitative Analysis of Meloxicam Metabolism

Sudoxicam (substrate), 5’-hydroxymethyl 5’-desmethyl meloxicam (metabolite standard), fluoxetine hydrochloride (internal standard), and 1,2-diamino-4,5-methylenedioxybenzene dihydrochloride (labeling agent) were obtained from Toronto Research Chemicals (Toronto, ON, Canada). Human liver microsomes pooled from 150 donors (HLM150) and recombinant enzyme Supersomes CYP1A2, 2A6, 2B6, 2C8, 2C9, 2C19, 2D6, 2E1, 3A4 and control prepared from baculovirus-transfected insect cells were purchased from Corning Gentest (Corning, NY).
