Insulin Signaling Pathway Modulation

Overexpression of NEDD4L in HUVECs

To detect the effect of overexpression of NEDD4L on HUVECs, we ordered the control and NEDD4L overexpressed adenovirus from HANBIO technology (Shanghai, China). Cells were infected with NEDD4L‐adenovirus and control adenovirus in ECM medium for 48 h. After infection, HUVECs were detected with tube formation assay, Transwell assay and immunostaining, as well as western blotting assay.
To examine the involved signalling pathway, cells were treated with Akt inhibitor MK‐2206, Erk1/2 inhibitor U0126 and eNOS inhibitor L‐NAME after being infected with adenovirus. Briefly, after being infected with these adenoviruses for 24 h, cells were treated with 100 nM MK‐2206 dihydrochloride (Cat#: HY‐10358, MedChemExpress, China), 20 μM U0126‐EtOH (Cat#: HY‐12031, MedChemExpress, China) and 100 μM L‐NAME hydrochloride (Cat#: HY‐18729A, MedChemExpress, China) for 24 h. Then, cells were collected for following detections, such as Matrigel tube formation assay, Transwell assay, western blotting and Phospho‐Histone H3 immunostaining.

Berberine and Autophagy Inhibition in Mice

The treatment group was given berberine (50 mg/kg, Med Chem Express, USA), intragastric administration once a day for 30 days. In addition to berberine, SIRT1 inhibition group was also given selisistat (20 mg/kg, Med Chem Express, USA) and was administered by intrabitoneal injection once every 3 days for 30 days. In addition to berberine, the mitochondria autophagy inhibition group was additionally given U0126-EtOH (10 mg/kg, Med Chem Express, USA) by intrabitoneal injection once every 3 days for 30 days. The mice were sacrificed to collect blood, gastrocnemius muscle, subcutaneous fat, visceral fat, and brown fat for subsequent use.

ERK inhibitor U0126-EtOH and p38 inhibitor SB203580 were purchased from MedChem Express (Monmouth Junction, NJ, USA), PE-anti-mouse CD117 (c-kit), APC-anti-mouse FcϵRIα, and FITC-anti-mouse IgE antibodies were purchased from Biolegend (San Diego, CA, USA). Recombinant interleukin (IL)-3 and stem cell factor (SCF) were purchased from PeproTech (Cranbury, NJ, USA). Evans blue and formamide were obtained from Dalian Meilun Biotechnology Co. Ltd. (Dalian, China). The ovalbumin (OVA; grade V) was purchased from Sigma Company (St. Louis, USA), and aluminum hydroxide was purchased from Thermo Fisher Scientific (Massachusetts, USA).
